生物信息数据库分析黏蛋白MUC2在结直肠癌中的表达及临床意义

摘　　要：目的:分析黏蛋白MUC2在结直肠癌中的表达水平,探究黏蛋白MUC2的表达与结直肠癌患者预后之间的关系。方法:利用TIMER2.0数据库探究黏蛋白MUC2在结直肠癌中的表达以及与免疫细胞浸润水平之间的相关性。通过GEPIA和Kaplan-Meier Plotter数据库分析黏蛋白MUC2对结直肠癌患者预后的影响。利用TISIDB数据库分析黏蛋白MUC2在结直肠癌不同免疫及分子亚型中的表达。结果:与配对正常结直肠组织相比,黏蛋白MUC2在结直肠癌组织中的表达明显降低,其表达与CD4^(+)T细胞呈正相关。黏蛋白MUC2高表达与结直肠癌患者较好的预后相关,且MUC2的表达与结直肠癌分子分型相关。结论:在结直肠癌中,黏蛋白MUC2具有潜在的抑癌作用,其表达减少参与肿瘤微环境中的免疫抑制。MUC2是一种潜在的结直肠癌预后分子标志物并有望成为结直肠癌靶向治疗的新靶点。Objective:To investigate the expression of MUC2 in colorectal cancer and its relationship with the prognosis of colorectal cancer patients.Methods:The expression of MUC2 in colorectal cancer and the correlation with the level of immune cell infiltration were explored by TIMER2.0 database.The effect of MUC2 on the prognosis of colorectal cancer patients were analyzed by GEPIA and KaplanMeier Plotter databases.The expression of MUC2 in different immune and molecular subtypes of colorectal cancer were examined by TISIDB database.Results:The expression of mucin MUC2 was significantly reduced in colorectal cancer tissues compared with paired normal colorectal tissues,and its expression was positively correlated with CD4^(+)T cells.The high expression of MUC2 was associated with better prognosis of colorectal cancer patients,and the expression of MUC2 was related to the molecular classification of colorectal cancer.Conclusion:MUC2 is a potential tumor suppressor in colorectal cancer,and its decreased expression is involved in immunosuppression in the tumor microenvironment.MUC2 is a potential prognostic molecular marker for colorectal cancer and is expected to become a new target for targeted therapy of colorectal cancer.
