摘  要:目的探讨不同剂量利塞膦酸钠对正畸大鼠牙根组织中FAK、基质金属蛋白酶2(MMP-2)、RANK蛋白表达及牙移动、牙根吸收的影响。方法选择120只SPF级的Wistar雄性大鼠,将120只大鼠按随机数字表法分为4组,包括模型组、利塞膦酸钠低剂量组、中剂量组、高剂量组,每组30只大鼠。高、中、低利塞膦酸钠的每日给药剂量为1、0.5、0.25 mg·kg^(-1)·d^(-1),模型组大鼠在相同时间给予0.9%氯化钠溶液灌胃。分析4组大鼠不同时间点的正畸牙移动距离、4组大鼠牙根组织形态、牙根组织中破骨细胞的数量、牙根组织中MMP-2、RANK及FAK蛋白表达。结果实验第7、14、21天,高剂量组的正畸牙移动距离、破骨细胞数量、明显较中、低剂量组、模型组大鼠低,中剂量明显较低剂量组、模型组低,低剂量组明显较模型组低,差异均有统计学意义(P<0.05)。苏木素和伊红染色后,实验第7天,模型组根分叉压力侧表面的牙骨质不连续,存在骨吸收陷窝,低、中剂量组也出现骨吸收陷窝,高剂量组骨吸收陷窝较浅;实验第14天,模型组的根分叉处、根尖处均有一定吸收,低、中剂量组牙根吸收多出现在受压力侧的根中1/3处,高剂量组大鼠根尖、根分叉形态基本完整,无明显的吸收;实验第21天,模型组的根尖受压力侧出现轻度吸收,低、中、高剂量组未出现根尖形态明显变化。实验第7、14、21天,高剂量组的牙根组织中MMP-2、RANK蛋白表达明显较中、低剂量组、模型组大鼠低,FAK蛋白明显高,中剂量组正畸牙根组织中MMP-2、RANK蛋白表达明显较低剂量组、模型组低,FAK蛋白明显高,低剂量组MMP-2、RANK蛋白表达明显较模型组低,FAK蛋白明显高,差异均有统计学意义(P<0.05)。结论利塞膦酸钠可抑制正畸大鼠的牙根吸收的影响,可能与抑制牙根组织中MMP-2、RANK蛋白表达,诱导FAK蛋白升高,抑制破骨细胞数量有关,且利塞膦酸钠剂量越�Objective To investigate the effects of different doses of risedronate on the expression of FAK,matrix metalloproteinase 2(MMP-2)and RANK proteins,tooth movement and root resorption in orthodontic rats.Methods A total of 120 SPF-grade Wistar male rats were randomly divided into 4 groups,including model group,low-dose risedronate group,medium-dose group and high-dose group,with 30 rats in each group.The daily dose of high,medium and low risedronate was 1,0.5 and 0.25 mg·kg^(-1)·d^(-1),and the rats in model group were given normal saline by gavage at the same time.The movement distance of orthodontic teeth,the morphology of root tissue,the number of osteoclasts in root tissue,and the expression of MMP-2,RANK and FAK protein in root tissue were analyzed.Results On the 7th,14th and 21st days of the experiment,the orthodontic tooth movement distance and the number of osteoclasts in the high-dose group were significantly lower than those in the middle and low dose groups and the model group,the middle dose group was significantly lower than those in the low dose group and the model group,and the low-dose group was significantly lower than that in the model group,the differences were statistically significant(P<0.05).After HE staining,on the 7th day of the experiment,the cementum on the pressure side of the root bifurcation in the model group was discontinuous and there were bone resorption lacunae.Bone resorption lacunae also appeared in the low and medium dose groups,and bone resorption lacunae were shallow in the high dose group.On the 14th day of the experiment,there was a certain amount of absorption at the root bifurcation and root tip in the model group,and the root resorption in the low-dose and medium-dose groups mostly occurred at 1/3 of the roots on the pressure side.The morphology of the root apex and root bifurcation in the high-dose group was basically intact without obvious absorption.On the 21d of the experiment,mild apical absorption was observed in the pressure side of the model group,while no signif
