摘 要:目的探究LncRNA FGFR3-AS1对卵巢癌(OC)的作用及其可能的机制。方法收集治疗的OC患者89例作为本次研究的患者组,另收集同期体检的正常人89例作为对照组。采用qRT-PCR检测患者组织与血清中FGFR3-AS1的相对表达,并分析FGFR3-AS1在OC患者中的临床意义与预后价值。通过体外实验分析FGFR3-AS1对OC细胞生物学功能的影响。结果FGFR3-AS1在OC组织和细胞中高表达(P<0.05),具有较好的临床诊断价值,且FGFR3-AS1高表达情况主要集中于Ⅲ+Ⅳ期,淋巴转移,低分化患者中(P<0.05)。COX分析发现FGFR3-AS1、TNM分期、淋巴转移是影响OC患者的独立预后因素。体外实验中,发现抑制FGFR3-AS1的表达可以抑制OC细胞的增殖、侵袭,促进细胞凋亡(P<0.05),而上调FGFR3-AS1的表达可以促进OC细胞的增殖、侵袭,抑制细胞凋亡。结论FGFR3-AS1在OC组织中存在高表达显著,可以促进OC细胞的活性,加速OC的发展,未来有望成为OC的潜在诊断和预后标志物。Objective To explore the effect of lncRNA FGFR3-AS1 on ovarian cancer(OC)and its possible mechanism.Methods A total of 89 OC patients treated in this hospital were collected as the patient group,and 89 healthy volunteers who received physical examinatiosn during the same period were collected as the control group.QRT-PCR was used to detect the relative expression of lncRNA FGFR3-AS1 in cancer tissues and serum samples.The clinical significance and prognostic value of lncRNA FGFR3-AS1 in OC were analyzed.The regulatory effects of lncRNA FGFR3-AS1 on the biological functions of OC cells were analyzed by in vitro experiments.Results LncRNA FGFR3-AS1 was upregulated in OC tissues and cells,with a good clinical diagnostic value.TOC patients with stageⅢ~Ⅳ,lymphatic metastasis,and poor differentiation expressed high levels of lncRNA FGFR3-AS1.COX analysis found that lncRNA FGFR3-AS1,TNM staging,and lymphatic metastasis were independent prognostic factors of OC.In vitro experiments showed that knockdown of lncRNA FGFR3-AS1 inhibited the proliferation and invasion of OC cells,and promoted cell apoptosis,and the opposite results were obtained after overexpression of lncRNA FGFR3-AS1.Conclusion LncRNA FGFR3-AS1 is highly expressed in OC tissues,which can promote cell activity and the development of OC.It is expected to become a potential diagnostic and prognostic marker of OC in the future.
