摘  要:目的探讨18β-甘草次酸对K19-C2mE自发胃肿瘤转基因鼠的干预效果及对环氧合酶-2(COX-2)、白介素-1β(IL-1β)表达的影响。方法选取40只状态良好的K19-C2mE转基因子代小鼠,随机分为模型组、干预组,另选择20只野生型C57BL/6N小鼠纳入对照组。模型组和对照组小鼠正常饮水,干预组小鼠6周龄时给予0.1%18β-甘草次酸水溶液作为饮水,3组小鼠每周称量体重,生长至52周时观察小鼠胃肿瘤情况,取小鼠胃黏膜组织,采用免疫组化方法检测微血管密度(MVD),采用蛋白印记杂交测定COX-2、IL-1β表达情况。结果3组小鼠各时间点体重差异无统计学意义(P>0.05)。对照组小鼠解剖可见胃黏膜形态光滑,未见肿瘤及其他病理表现;模型组及干预组小鼠可见胃内肿物形成,伴有胃黏膜出血;模型组小鼠胃肿瘤发生率显著高于干预组(P<0.05)。HE染色显示,对照组小鼠胃组织未见增生性病变,模型组及干预组小鼠可见慢性炎症,淋巴滤泡形成及增生性病变。模型组小鼠黏膜MVD及COX-2、IL-1β表达量显著高于对照组及干预组(P<0.05),干预组显著低于模型组(P<0.05)。结论18β-甘草次酸可降低K19-C2mE自发胃肿瘤转基因鼠胃肿瘤发生率,减轻黏膜炎性反应,抑制新生血管生成,其机制可能与下调COX-2、IL-1β等因子表达有关。Objective To explore intervention effects of 18β-glycyrrhetinic acid on K19C2mE transgenic mice with spontaneous gastric tumor and corresponding influences on expressions of cyclooxygenase-2(COX-2)and interleukin-1β(IL-1β).Methods Forty K19-C2mE transgenic offspring mice in good conditions were selected and randomly divided into the model group and intervention group.Another 20 wild-type C57BL/6N mice were included in the control group.Mice in the model group and the control group were given drank water,mice in the intervention group were administered with 0.1%18β-glycyrrhetinic acid aqueous solution as drinking water when they were 6 weeks old.Mice in each group were weighted weekly,gastric tumors were observed as 52 weeks old,gastric mucosa tissues were taken to test the microvessel density(MVD)by immunohistochemical method,and the expressions of COX-2 and IL-1βwere determined by Western blot.Results Difference in weight at time points weren’t statistically significant between groups(P>0.05).Smooth gastric mucosa without tumors or other pathological features emerged in the control group.Intra-gastric tumors accompanied by gastric mucosal hemorrhage were found in the model group and the intervention group.The incidence of gastric tumors in the model group was significantly higher than that in the intervention group(P<0.05).Hematoxylin and eosin(H&E)staining showed no proliferative lesions were found in gastric tissues in the control group.Chronic inflammation,lymphoid follicle formation and proliferative lesions were noted in the model group and the intervention group.MVD,COX-2 and IL-1βwere significantly higher in the model group than in the control group and the intervention group(P<0.05),which were significantly lower in the intervention group than in the model group(P<0.05).Conclusion The 18β-glycyrrhetinic acid is capable of reducing the incidence of gastric tumors in K19-C2mE transgenic mice,relieving mucosal inflammation,and inhibiting angiogenesis.Corresponding mechanism may be related to down-r
