摘 要:目的探究管家基因GAPDH能否有效诊断肝癌患者并影响其生存预后。方法利用TCGA、GEO等生物信息学数据库,检测GAPDH在肝癌中的表达;利用K-M plotter数据库,验证GAPDH与肝癌患者生存预后的关联;利用DAVID、TIMER数据库,探究GAPDH影响患者生存预后的相关机制。结果GAPDH在肝癌中表达升高,是一个影响肝癌患者总生存期和无复发生存期的独立危险因素。GAPDH可能通过调控糖酵解过程、缺氧诱导因子-1相关信号通路和免疫细胞浸润来影响患者生存率。结论GAPDH是一个潜在的肝癌诊断指标和预后影响因素。Objective To explore whether the housekeeping gene GAPDH can be used to effectively diagnose liver cancer patients and affect their survival prognosis.Methods The expression of GAPDH in hepatocellular carcinoma was detected by using a variety of bioinformatics databases,including TCGA and GEO;K-M plotter database was used to verify the association between GAPDH and survival and prognosis of patients with liver cancer;DAVID and TIMER databases were used to explore the mechanism of GAPDH affecting the survival and prognosis of patients.Results The increased expression of GAPDH in hepatocellular carcinoma was showed to be an independent risk factor affecting the overall survival and recurrence free survival of patients with hepatocellular carcinoma.GAPDH could affect the survival rate of patients by regulating glycolysis,hypoxia inducible factor-1 related signal pathway and immune cell infiltration.Conclusion GAPDH could serve as a potential diagnostic index and prognostic factor for hepatocellular carcinoma.
