摘 要:目的 探讨高迁移率族蛋白A2(HMGA2)和叉头转录因子O亚型3a(FOXO3a)在胃癌组织中的表达及其临床意义。方法 选择45例胃癌组织标本为研究组,以其癌旁组织为对照组,采用实时荧光定量PCR法检测胃癌组织及癌旁组织HMGA2 mRNA和FOXO3a mRNA,采用免疫组化法检测胃癌组织及癌旁组织中的HMGA2和FOXO3a,用SPSS 19.0软件对结果进行统计学分析。结果 HMGA2 mRNA在胃癌组织中表达水平显著高于癌旁组织,FOXO3a mRNA表达水平显著低于癌旁组织(P<0.05)。HMGA2在胃癌组织中阳性率显著高于癌旁组织,FOXO3a阳性率显著低于癌旁组织(P<0.05)。TNM分期越高、分化程度越低、有淋巴结转移的HMGA2表达越高,FOXO3a表达越低(P<0.05)。HMGA2和FOXO3a成负相关(r=-0.435,P=0.000)。Kaplan-Meier生存分析显示,HMGA2低表达组比高表达组有更好的累积生存率,FOXO3a高表达组比低表达组有更好的累积生存率(P<0.05)。结论 HMGA2在胃癌组织中表达水平高于癌旁组织、FOXO3a表达水平低于癌旁组织,其异常表达可作为胃癌诊断的分子标记物。Objective This paper aims to investigate the expression and clinical significance of high mobility group box A2(HMGA2) and forkhead transcription factor O isoform 3a(FOXO3a) in gastric cancer tissues. Methods A total of 45 gastric cancer tissue specimens were selected as the research group, and their adjacent tissues were used as the control group. Real-time fluorescence quantitative PCR was used to detect HMGA2 mRNA and FOXO3a mRNA in gastric cancer tissues and adjacent tissues, and immunohistochemistry was used to detect HMGA2 and FOXO3a in gastric cancer tissues and adjacent tissues. SPSS 19.0 software was used for statistical analysis. Results The expression of HMGA2 mRNA in gastric cancer tissue was significantly higher than that in adjacent tissue, and the expression of FOXO3a mRNA was significantly lower than that in adjacent tissue(P<0.05). The positive rate of HMGA2 in gastric cancer tissue was significantly higher than that in adjacent tissue, and the positive rate of FOXO3a was significantly lower than that in adjacent tissue(P<0.05). The higher the TNM stage, the lower the degree of differentiation, the higher the expression of HMGA2 with lymph node metastasis, and the lower the expression of FOXO3a(P<0.05). HMGA2 and FOXO3a were negatively correlated(r=-0.435, P=0.000). Kaplan-Meier survival analysis showed that the HMGA2 low expression group had a better cumulative survival rate than the high expression group, and the FOXO3a high expression group had a better cumulative survival rate than the low expression group(P<0.05). Conclusion The expression of HMGA2 in gastric cancer tissue is higher than that in adjacent tissue, and the expression of FOXO3a is lower than that in adjacent tissue. Its abnormal expression can be used as a molecular marker for the diagnosis of gastric cancer.
