PHAP1和SUMO2在宫颈鳞状细胞癌中的表达及其临床病理学意义

摘　　要：目的探讨宫颈鳞状细胞癌(CSCC)中PHAP1及SUMO2的表达,分析其临床病理意义。方法采用RT-PCR法及IHC-SP法检测组织中PHAP1、SUMO2的表达。运用WB法检测PHAP1及SUMO2在Ect1、C33A和SiHa细胞中的表达。结果RT-PCR结果显示,PHAP1(9.22±8.45)及SUMO2(1.82±2.70)在CSCC组织中高表达,PHAP1在CSCC组织与正常宫颈组织间差异有统计学意义(P=0.007)。IHC结果显示,PHAP1及SUMO2在正常组织、CIN组织、CSCC组织内的表达呈递增趋势,且各组间差异均有统计学意义;SUMO2高表达与不同FIGO分期(P=0.024)和淋巴结转移(P=0.020)患者SUMO2表达差异有统计学意义。PHAP1及SUMO2蛋白在Ect1、C33A和SiHa细胞的表达呈增高趋势,且SiHa与Ect1(P=0.002)、C33A与Ect1(P=0.008)间表达差异有统计学意义。结论PHAP1及SUMO2在宫颈癌中呈高表达,其过度表达可能促进宫颈鳞癌的发生和转移;PHAP1和SUMO2在宫颈鳞癌中的高表达与HPV16感染无显著相关性。Objective To investigate the expression of PHAP1 and SUMO2 in cervical squamous cell carcinoma(CSCC)and to analyze the clinicopathological significance of CSCC.Methods The expression levels of PHAP1,SUMO2 were detected by RT-PCR and IHC-SP.The expression of PHAP1 and SUMO2 in Ect1,C33A and SiHa cell lines was detected by Western blot.Results RT-PCR showed that the expression of PHAP1(9.22±8.45)and SUMO2(1.82±2.70)increased in CSCC group.There was statistical significance of PHAP1 between the tumor group and the normal group(P=0.007).IHC results showed that the positive rates of PHAP1 and SUMO2 in CSCC,CIN group,and in normal tissues were increasing,and there was statistical significance among all groups.The high expression of SUMO2 was related to FIGO stage(P=0.024)and lymph node metastasis(P=0.020),the expression of PHAP1 was not associated with any clinical parameters.The expression of PHAP1 and SUMO2 proteins showed an increasing trend in Ect1,C33A and SiHa cell lines,and there were statistically significant differences between SiHa and Ect1(P=0.002),C33A and Ect1(P=0.008).Conclusion The PHAP1 and SUMO2 are highly expressed in cervical carcinoma,and their overexpression may promote the occurrence of cervical squamous cell carcinoma.The high expression of PHAP1 and SUMO2 in CSCC is not significantly correlated with HPV16 infection.
