摘  要:旨在探究GDF8基因(Growth differentiation factor 8)编码蛋白的结构和功能,对后期研究其生理机制提供理论依据。利用生物信息学技术对牛GDF8蛋白二级和三级结构进行了预测,分析了保守结构域、信号肽剪切位点、理化性质、亲疏水性、亚细胞定位和蛋白网络通路。结果显示,牛GDF8蛋白与人和小鼠具有相似的保守结构域,不稳定指数85.52,属于不稳定蛋白,其亲水性氨基酸明显多于疏水性氨基酸,为亲水性蛋白;牛GDF8蛋白在内质网分布的概率44.4%,在线粒体分布的概率33.3%,二级结构以无规则卷曲为主,折叠缠绕后形成三级结构;牛GDF8蛋白PPI网络图有11个节点,其KEGG通路主要与骨骼肌纤维发育、活化素受体信号通路、成肌细胞融合等相关。综上,推测GDF8蛋白存在于内质网和线粒体中,通过与亲水性蛋白结合调节机体生物学功能。The purpose of this study was to explore the structure and function of GDF8 encoding protein,and provide theoretical basis for studying physiological mechanisms of GDF8.Bioinformatics techniques were used to predict GDF8 protein secondary and tertiary structure,and analyze the conserved domain,signal peptide splice site,physicochemical properties,hydrophilicity,subcellular localization and PPI network.The results showed that bovine GDF8 protein has similar domain with human and mouse.The instability index of GDF8 protein was 85.52,and it was hydrophilic protein because of hydrophilic amino acids were significantly more than hydrophobic amino acids.The probability of distribution in endoplasmic reticulum was 44.4%,and the distribution probability in mitochondria was 33.3%.The secondary structure of GDF8 protein was mainly formed from random coil,and the tertiary structure was formed after its folding and winding.PPI network of bovine GDF8 protein had 11 nodes,and KEGG pathway was mainly related to skeletal muscle myofiber development,activin receptor signaling pathway,and myoblast fusion.In summary,it is speculated that GDF8 protein exists in endoplasmic reticulum and mitochondria,and regulates biological function by binding with hydrophilic proteins.
