摘　　要：目的 探讨MOB3B在结直肠癌中的表达及其与生存预后的关系。方法 通过数据库分析MOB3B在结直肠癌组织与癌旁正常组织中的表达及其与生存预后的关系。通过免疫组化、qPCR实验检测结直肠癌组织及其癌旁正常组织中MOB3B的表达情况,以及通过qPCR实验检测结直肠癌细胞(RKO、HT29、HCT116、SW480、SW620、DLD1、Caco2)和人正常结直肠黏膜细胞(FHC)中MOB3B mRNA的表达。结果 GEO数据库的4个数据集的分析结果显示,MOB3B在结直肠癌组织中的表达均低于癌旁正常组织(均P<0.05)。GEPIA在线分析结果显示,MOB3B在结肠癌组织、直肠癌组织中的表达均低于各对应癌旁正常组织(均P<0.05)。OS、DFS生存曲线结果显示,在结直肠癌中,MOB3B相对低表达的患者的OS、DFS均短于MOB3B相对高表达的患者(均P<0.05)。免疫组化实验结果显示,MOB3B在结直肠癌组织中的表达低于癌旁正常组织(P<0.05)。qPCR实验结果显示,MOB3B mRNA在结直肠癌组织中的表达低于癌旁正常组织,在结直肠癌细胞中的表达均低于人正常结直肠黏膜细胞(均P<0.05)。结论 MOB3B在结直肠癌中低表达,且其低表达可能与结直肠癌的不良预后相关。Objectives To investigate the expression of MOB3B in colorectal cancer and its relationship with survival prognosis.Methods The expression of MOB3B in colorectal cancer and adjacent normal tissues, and its relationship with survival prognosis was analyzed based on database information. The expression of MOB3B in colorectal cancer and adjacent normal tissues was detected by immunohistochemistry and qPCR. The expression of MOB3B mRNA in colorectal cancer cells(RKO, HT29, HCT116,SW480, SW620, DLD1, Caco2) and human normal colorectal mucosal cells(FHC) was detected by qPCR. Results The analysis results of the four datasets in GEO database showed that the expression of MOB3B in colorectal cancer tissues was significantly lower than that in adjacent normal tissues(P<0.05). The results of GEPIA online analysis showed that the expression of MOB3B in colon cancer tissues and rectal cancer tissues were significantly lower than that in corresponding adjacent normal tissues(P<0.05). The OS and DFS of patients with low expression of MOB3B were significantly shorter than those of patients with high expression of MOB3B(P<0.05). The expression of MOB3B protein in colorectal cancer tissues was significantly lower than that in adjacent normal tissues(P<0.05). The expression of MOB3B m RNA in colorectal cancer tissues was significantly lower than that in adjacent normal tissues, and the expression of MOB3B mRNA in colorectal cancer cells was significantly lower than that in human normal colorectal mucosal cells(P<0.05). Conclusion The expression of MOB3B is low in colorectal cancer, and its low expression may be related to poor prognosis.
