摘 要:目的研究美丽芍药Paeonia mairei Levl.的化学成分和生物活性研究。方法美丽芍药70%乙醇提取物采用柱色谱进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。对其中成分进行体外激动褪黑素受体1活性、抗炎和细胞毒性、抑制黄嘌呤氧化酶活性研究。结果从中分离得到11个化合物,分别鉴定为芍药苷(1)、氧化芍药苷(2)、没食子酰芍药苷(3)、芍药内酯苷(4)、6-O-benzoyl paeoniflorin(5)、benzoyl oxypaeoniflorin(6)、mudanpioside C(7)、1,2,3,4,6-五没食子酰葡萄糖(8)、邻苯三酚(9)、蔗糖(10)、paeonidanin E(11)。化合物1~8在100μmol/L浓度下对褪黑素受体1均无活性,在5~100μmol/L浓度范围内无细胞毒作用和抗炎活性。在200μmol/L浓度作用下,五没食子酰葡萄糖和没食子酰芍药苷有一定的黄嘌呤氧化酶抑制活性,该酶活率分别为(65.98±3.46)%、(74.17±2.13)%。结论化合物2~11为首次从该植物中分离得到。AIM To study the chemical constituents from Paeonia mairei Levl.and their biological activities.METHODS The 70%ethanol extract from P.mairei was isolated and purified by column chromatography,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Compounds were bio-assayed on the agitating human melatonin receptor 1(MT1),anti-inflammatory and cytotoxic activity,inhibition of xanthine oxidase activity in vitro.RESULTS Eleven compounds were isolated and identified as paeoniflorin(1),oxypaeoniflorin(2),galloylpaeoniflorin(3),albiflorin(4),6-O-benzoyl paeoniflorin(5),benzoyl oxypaeoniflorin(6),mudanpioside C(7),pentagalloyglucose(8),pyogallol(9),sucrose(10),paeonidanin E(11).Compounds 1-8 showed no agitating activity on MT1 receptor at the concentration of 100μmol/L and no cytotoxic or anti-inflammatory activities at concentration of 5 to 100μmol/L.At the concentration of 200μmol/L,pentagalloyglucose and galloylpaeoniflorin showed moderate xanthine oxidase inhibitory activities,which enzymatic activites decreased to(65.98±3.46)%and(74.17±2.13)%.CONCLUSION Compounds 2-11 are isolated from this plant for the first time.
