摘 要:目的探讨梓醇-川芎嗪方对阿尔茨海默病(AD)小鼠学习记忆行为及相关炎性因子的影响。方法将50只SPF级APPswe/PS1ΔE9双转基因雄性小鼠随机分为模型组、安理申组及梓醇-川芎嗪方低、中、高剂量组,每组10只;选取同月龄C57BL/6J野生型雄性小鼠10只作为正常组。梓醇-川芎嗪方低、中、高剂量组分别给予25 mg/kg、50 mg/kg、100 mg/kg梓醇-川芎嗪方灌胃,安理申组给予安理申溶液5 mg/kg灌胃,正常组、模型组给予等体积生理盐水灌胃,均每天1次,连续灌胃8周。Morris水迷宫实验检测记录第1~5天小鼠逃避潜伏期和第6天的穿越原平台次数,新物体识别测试小鼠1 h、24 h对新物体的偏好指数,酶联免疫吸附法(ELISA)检测各组小鼠血清和脑组织海马区神经相关蛋白[β淀粉样蛋白(Aβ)、Tau蛋白、磷酸化Tau蛋白(p-Tau)]含量和炎性因子[白细胞介素-4(IL-4)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)]水平。结果第3~5天,梓醇-川芎嗪方各组和安理申组小鼠的逃避潜伏期均明显短于模型组(P均<0.05);梓醇-川芎嗪方高剂量组第4天、第5天的逃避潜伏期均明显短于川芎嗪方中剂量组和安理申组(P均<0.05);梓醇-川芎嗪方各组和安理申组小鼠穿越原平台次数均明显多于模型组(P均<0.05),且梓醇-川芎嗪方高剂量组明显多于梓醇-川芎嗪方低、中剂量组及安理申组(P均<0.05)。梓醇-川芎嗪方各组和安理申组1 h、24 h对新物体的偏好指数均明显高于模型组(P均<0.05),且梓醇-川芎嗪方高剂量组明显高于梓醇-川芎嗪方低、中剂量组及安理申组(P均<0.05),梓醇-川芎嗪方中剂量组及安理申组均明显高于梓醇-川芎嗪方低剂量组(P均<0.05)。梓醇-川芎嗪方各组和安理申组小鼠血清和脑组织海马区Aβ、Tau、p-Tau蛋白含量和IL-4、IL-6、IL-10、TNF-α水平均明显低于模型组(P均<0.05),且梓醇-川芎嗪方高剂量组明显�Objective It is to investigate the effects of catalpol-tetramethylpyrazine prescription(CT)on learning and memory behavior and related inflammatory factors in Alzheimer’s disease(AD)model mice.Methods Fifty SPF APPswe/PS1ΔE9 double transgenic male mice were randomly divided into model group,Aricept group,CT low dose(CT-L)group,CT medium dose(CT-M)group,CT high dose(CT-H)group.10 C57BL/6J male mice with the same months old were taken as normal group.The CT-L group,CT-M group and CT-H group were given low-dose,medium-dose and high-dose CT by gavage respectively,and the Aricept group was given Aricept solution by gavage.The normal group and model group were given the same volume of normal saline by gavage.All once per day,continuously treated for 8 weeks.The escape latency from 1 to 5 days and number of crossing platforms on the 6th day of the mice were detected by Morris water maze test,the preference index for new objects in 1 h and 24 h of the mice was detected by new object recognition test,The contents of neural-associated proteins[β-amyloid(Aβ),Tau protein,phosphorylated Tau protein(p-Tau)]and levels of inflammatory factors[interleukin-4(IL-4),interleukin-6(IL-6),interleukin-10(IL-10),tumor necrosis factor-α(TNF-α)]in serum and hippocampal region of brain tissue of the mice in each group were measured by enzyme-linked immunosorbent assay(ELISA).Results On the 3rd~5th day,the escape latency of the mice in each CT group and the Anrishen group was significantly shorter than that of the model group(all P<0.05);the escape latency of the mice in the CT high-dose group on the 4th and 5 th days was significantly shorter than that of the CT medium-dose group and the Anrishen group(all P<0.05);the number of crossing platform of the mice in each CT group and the Anrishen group was significantly more than that of the model group(all P<0.05),the number of crossing platform of the mice in the CT high-dose group was significantly more than that of the CT low-dose group,CT middle-dose group and the Anrishen group(all P
