摘 要:目的:观察白芍多酚对急性酒精性肝损伤小鼠的防护作用,并探讨可能机制。方法:取40只ICR小鼠,随机分为健康组、肝损伤组、白芍多酚组、联合组,每组10只。联合组灌胃白芍多酚(100 mg/kg),腹膜内注射ML385[核因子E2相关因子2(Nrf2)/Kelch样环氧氯丙烷相关蛋白1(Keap1)信号通路抑制剂](30 mg/kg);白芍多酚组灌胃白芍多酚(100 mg/kg),腹膜内注射等量DMSO;健康组、肝损伤组灌胃等体积生理盐水,腹膜内注射等量DMSO。每日1次,干预7 d。肝损伤组、白芍多酚组、联合组一次性灌胃50%乙醇溶液(12 ml/kg)建立急性酒精性肝损伤模型。检测各组小鼠肝脏指数、血清肝功能指标、血清氧化应激指标;Tunel染色检测肝细胞凋亡;Western blot法检测肝组织Nrf2、Keap1蛋白表达。结果:与肝损伤组比较,白芍多酚组小鼠肝脏指数、血清谷草转氨酶(AST)、丙氨酸氨基转移酶(ALT)、丙二醛(MDA)水平、肝细胞凋亡率、肝组织Nrf2蛋白表达降低,血清超氧化物歧化酶(SOD)活性、肝组织Keap1蛋白表达升高(P<0.05);与白芍多酚组比较,联合组小鼠肝脏指数、血清AST、ALT、MDA水平、肝细胞凋亡率、肝组织Nrf2蛋白表达升高,血清SOD活性、肝组织Keap1蛋白表达降低(P<0.05)。结论:白芍多酚可防止肝损伤、保护肝功能、抑制氧化应激及肝细胞凋亡,推测其作用机制与激活Nrf2/Keap1信号通路有关。Objective:To observe the protective effect of white peony polyphenols on acute alcoholic liver injury in mice,and to explore the possible mechanism.Methods:Take 40 ICR mice and randomly divide them into the healthy group,liver injury group,white peony poly-phenol group,and combination group,ten mice in each group.Mice in the combination group were intragastrically administered with white peony polyphenols (100 mg/kg) and injected intraperitoneally with ML385[nuclear factor E2-related factor 2 (Nrf2)/Kelch-like epichloro-hydrin-related protein 1 (Keap1) signaling pathway inhibitor](30 mg/kg).Mice in the white peony polyphenol group were given intraper-itoneal injections of the same amount of DMSO with white peony polyphenol (100 mg/kg).The mice in the healthy group and the liver injury group were given an equal volume of normal saline and injected intraperitoneally with an equal volume of DMSO.One time a day,intervention for seven days.Acute alcoholic liver injury models were established in the liver injury group,the white peony polyphenol group,and the com-bination group.The liver index was detected;serum liver function index was detected;serum oxidative stress index was detected;Tunel stai-ning was used to detect hepatocyte apoptosis;Western blot method was used to detect the expression of Nrf2 and Keap1 protein in liver tissue.Results:Compared with the liver injury group,the liver index,serum aspartate aminotransferase (AST),alanine aminotransferase (ALT),malondialdehyde (MDA) levels,liver cell apoptosis rate,liver tissue Nrf2 protein expression were decreased,serum superoxide dismutase(SOD) activity,and liver tissue Keap1 protein expression was increased in the white peony polyphenol group (P<0.05).Compared with the white peony poly phenol group,the liver index,serum AST,ALT,MDA levels,liver Apoptosis rate,liver tissue Nrf2 protein expression were increased,serum SOD activity,liver tissue Keap1 protein expression were decreased in the combined group (P<0.05).Conclusion:White peony polyphenols can treat a liver
