摘 要:以山楂叶为研究对象,研究其总黄酮提取工艺及降脂活性。在单因素试验基础上,采用Box-Behnken响应面法对山楂叶总黄酮的提取工艺进行优化,考察乙醇体积分数、液料比和提取时间对其总黄酮产率的影响,以胰脂肪酶抑制率为指标评价山楂叶总黄酮的降脂活性。结果表明,最佳提取条件为乙醇体积分数56%、液料比28︰1(mL/g)、提取时间34 min,此时山楂叶总黄酮产率为4.43%。活性测定结果表明,山楂叶总黄酮具有较好的降脂活性,且具有一定的浓度依赖性。优选的山楂叶总黄酮提取工艺方法可行,其总黄酮具有一定降脂活性。Optimize the extraction process and evaluate hypolipidemic effects of total flavonoids in the hawthorn leaves.Based on a single factor test,the ethanol concentration,the ratio of solvent to material and the extraction time were used as the factors,and the Box-Behnken was used to design the extraction process.The inhibition rate of pancreatic lipase was used to assess the hypolipidemic effect of total flavonoids in the hawthorn leaves.The result showed that the optimal extraction condition was as follows:ethanol concentration,the ratio of solvent to material and extraction time were 56%,28︰1(mL/g),and 34 min,respectively.Under the above condition,the experimental yield of total flavonoids in the hawthorn leaves was 4.43%.Furthermore,total flavonoids in the hawthorn leaves exhibited significant hypolipidemic effects in vitro in a dosedependent manner.The optimum extraction process of the flavonoids from the hawthorn leaves is feasible,and total flavonoids in the hawthorn leaves have significant hypolipidemic effects.
