摘  要:酶联免疫吸附法在众多领域被广范应用,但因其影响因素较多以及重演性问题的残留,导致酶联免疫吸附检测标准化程度较低。为推动酶联免疫吸附检测标准化进程,研究了酶联免疫吸附法检测花生中黄曲霉毒素B_(1)的过程中封闭条件、标准品稀释液中甲醇浓度、pH值等影响因素对实验结果的影响程度。研究结果表明,缓冲溶液pH值以及反应体系盐离子浓度对实验影响程度较大,可作为推进标准化进程的关键突破点,此外其他影响因素的研究为提高实验安全性和降低检测成本提供了一定数据支撑。研究建立了黄曲霉毒素B_(1)空白标准曲线和花生基质标准曲线,标准曲线检测范围为(IC 20~IC 80)0.001~0.027 ng/mL,利用加标回收实验测定的回收率为88.1%~95.1%。Enzyme-linked immunosorbent assay(ELISA)has been widely used in many fields,but the standardization degree of ELISA is low due to many influencing factors and reproducibility problems.In order to promote the standardization of ELISA,the influence degree of methanol concentration,pH and other influencing factors on the experimental results of aflatoxin B_(1) in peanut was studied in the process of ELISA.The pH of buffer solution and the concentration of salt ions in the reaction system have great influence on the experiment,which can be used as a key breakthrough point to promote the standardization process.In addition,the study on other influencing factors provides certain data support for improving the experimental safety and reducing the detection cost.Meanwhile,the blank standard curve of aflatoxin B_(1) and peanut matrix standard curve were established.The detection range of standard curve was(IC 20-IC 80)0.001~0.027 ng/mL,and the recovery rate was 88.1%~95.1%by spiked recovery experiment.
