摘 要:目的探讨环状RNA(circRNA,Circ)_101237调控肝癌细胞进展的分子机制。方法将新乡医学院第一附属医院2019年7月至2021年7月行手术切除的肝癌组织和癌旁组织作为研究对象,采用荧光定量聚合酶链反应(PCR)分析癌旁组织和肝癌组织Circ_101237表达水平。采用sh-Con和sh-Circ_101237慢病毒感染人肝癌细胞系MHCC97H,建立sh-Con组和sh-Circ_101237组细胞系,细胞计数试剂盒(CCK-8)、克隆形成实验、划痕实验和Transwell实验分析Circ_101237对细胞增殖、迁移和侵袭能力的影响。双荧光素酶报告实验分析Circ_101237的靶基因。组间数据比较采用t检验。结果癌旁组织中Circ_101237表达水平(1.01±0.18)明显低于肝癌组织Circ_101237表达水平(2.09±0.26),差异有统计学意义(t=30.560,P<0.05)。sh-Con组细胞吸光度值(A)、细胞克隆形成数量、划痕愈合率和侵袭数量[(1.97±0.09),(164.67±14.40)个,(62.50±5.51)%,(130.83±25.18)个]明显高于sh-Circ_101237组细胞A值\细胞克隆形成数量、划痕愈合率和侵袭数量[(1.33±0.18),(111.33±9.07)个;(28.63±6.84)%,(92.33±14.05)个],差异有统计学意义(t=7.782、7.675、9.444、3.270,P<0.05)。Circ_101237与miR-490-3p存在连续互补结合位点。sh-Con组细胞miR-490-3p表达水平(0.99±0.14)明显低于sh-Circ_101237组细胞miR-490-3p表达水平(2.17±0.24),差异有统计学意义(t=10.280,P<0.05)。miR-NC+sh-Circ_101237组细胞A值(1.01±0.13)明显低于miR-490-3p抑制剂+sh-Circ_101237组细胞A值(1.60±0.09),差异有统计学意义(t=9.143,P<0.05)。miR-NC+sh-Circ_101237组细胞划痕愈合率[(27.41±5.15)%]明显低于miR-490-3p抑制剂+sh-Circ_101237组划痕愈合率[(49.19±7.81)%],差异有统计学意义(t=5.703,P<0.05)。miR-NC+sh-Circ_101237组细胞侵袭数量[(90.83±9.87)个]明显低于miR-490-3p抑制剂+sh-Circ_101237组细胞侵袭数量[(122.33±11.09)个],差异有统计学意义(t=5.197,P<0.05)。结论Circ_101237在肝癌组织中呈高表达,通过miR-490-3p调节�Objective To explore the molecular mechanism of circular RNA(Circ)_101237 regulating the progression of hepatocellular carcinoma(HCC)cells.Methods The liver cancer tissues and adjacent tissues resected in the First Affiliated Hospital of Xinxiang Medical University from July 2019 to July 2021 were selected as research objects,and the Circ_101237 of adjacent tissues and liver cancer tissues was analyzed by fluorescent quantitative polymerase chain reaction(PCR).Human HCC cell line MHCC97H was infected by sh-Con and sh-Circ_101237 lentivirus to establish sh-Con group and sh-Circ_101237 group respectively.Cell proliferation,migration and invasion were analyzed by cell counting kit-8(CCK-8)assay,cloning formation test,scratch test and Transwell test.The target gene of Circ_101237 was analyzed by double Luciferase Reporting Experiment.T-test was used to compare the data between groups.Results The expression level of Circ_101237 in the adjacent tissues(1.01±0.18)was significantly lower than that of HCC(2.09±0.26,t=30.560,P<0.05).The absorbance(A)value,number of cell clones,scratch healing rate and number of invasive cells in sh-Con group[(1.97±0.09),(164.67±14.40)cells,(62.50±5.51)%,(130.83±25.18)cells]were significantly increased as compared with than those in sh-Circ group[(1.33±0.18),(111.33±9.07)cells;(28.63±6.84)%,(92.33±14.05)cells,t=7.782,7.675,9.444,3.270,P<0.05].The miR-490-3p was target gene of Circ_101237.The expression level of miR-490-3p in sh-Con group(0.99±0.14)was significantly lower than that of sh-Circ_101237 group(2.17±0.24,t=10.280,P<0.05).The A value of cells in miR-NC+sh-Circ_101237 group(1.01±0.13)was significantly lower than that in the miR-490-3p inhibitor+sh-Circ_101237 group(1.60±0.09,t=9.143,P<0.05).The wounding healing rate in miR-NC+sh-Circ_101237 group[(27.41±5.15)%]was significantly lower than that in miR-490-3p inhibitor+sh-Circ_101237 group[(49.19±7.81)%,t=5.703,P<0.05].The number of invasive cells in the miR-NC+sh-Circ_101237 group[(90.83±9.87)cells]was significantly
