摘 要:目的 优化东北刺人参总皂苷(Oplopanax elatus Nakai. extract, ONE)提取工艺,并考察其对果蝇睡眠-觉醒节律的调节作用。方法 采用乙醇回流法,以总皂苷得率为考察指标,在单因素试验基础上,利用Box-Behnken响应曲面法优化ONE的最佳提取工艺。以果蝇为考察对象,人参提取物(ginseng extract,GE)为对照,考察ONE的镇静、改善睡眠作用和对睡眠-觉醒节律的影响。结果 优化后ONE的最佳提取工艺为乙醇体积分数70%、液固比30:1 (mL:g)、提取温度90℃、提取时间60 min,总皂苷得率为(3.08±0.09)%。药理作用研究显示,不同于GE对中枢神经系统低剂量兴奋、高剂量抑制的作用特点, ONE可以减少果蝇的自主活动,延长夜间睡眠时间,改善睡眠-觉醒节律。结论 优化的提取工艺和药理作用可为东北刺人参的产品开发和临床应用提供理论依据。Objective To optimize the extraction process of total saponins from Oplopanax elatus Nakai. extract(NOE) and investigate its regulatory effects on the sleep-wake rhythm of Drosophila melanogaster. Methods On the basis of a single-factor experiment, the Box-Behnken response surface methodology was used to optimize the extraction process of ONE by ethanol reflux method. With Drosophila melanogaster as an experimental object and ginseng extract(GE) as a control, the sedative effects, sleep improvement, and sleep-wake rhythm of ONE was studied. Results The optimum extraction conditions of ONE were ethanol volume fraction 70%, a liquid-solid ratio of 30:1(mL:g), extraction temperature of 90℃, and extraction time of 60 min. The yield of total saponins was(3.08±0.09)%. Pharmacological studies had shown that different from the characteristics of low-dose excitation and high-dose inhibition of GE on the central nervous system, ONE could reduce the autonomous activity of Drosophila melanogaster, prolong nighttime sleep, and improve sleep-wake rhythm. Conclusion The optimized extraction process and pharmacological action can provide a theoretical basis for the product development and clinical application of Oplopanax elatus Nakai..
