摘 要:通过缓冻协同微波法提取竹荪多糖,以20%~80%乙醇分级制备得到4种多糖组分,分别为PI-20、PI-40、PI-60和PI-80;采用UV、FI-IR、HPSEC-MALLS-RI、HPAEC对多糖结构进行分析.结果表明4种组分均含有少量蛋白质,但未检测到糖醛酸存在,PI-20和PI-40多糖含量均在50%以上;低浓度乙醇醇沉的多糖PI-20和PI-40的相对分子质量达到千万级,而高体积分数乙醇醇沉的多糖相对分子质量在万级水平;20%~60%醇沉多糖均由Gal、Glu和Man构成,PI-80仅由Glu和Man组成.4种醇沉多糖组分均具有一定的抗氧化能力,在3种抗氧化试验中,4种醇沉组分PI-80综合抗氧化能力最佳,清除ABTS自由基半抑制质量浓度为0.2484 mg/mL,PI-40综合抗氧化活性最弱,清除ABTS自由基半抑制质量浓度为2.9043 mg/mL.The polysaccharide of Dictyophora indusiata was extracted by slow freezing and microwave method.Four polysaccharide components,PI-20,PI-40,PI-60 and PI-80,were prepared respectively by fractionation with 20%~80%ethanol.UV,FI-IR,HPSEC-MALLS-RI,HPAEC were used to analyze the structure of polysaccharide.The results showed that the four components contained a small amount of protein,but no uronic acid was detected,and the content of PI-20 and PI-40 polysaccharides was more than 50%.The molecular weight of PI-20 and PI-40 precipitated by low concentration ethanol reached tens of millions,while the molecular weight of polysaccharide precipitated by high concentration ethanol reached ten thousand.20%~60%ethanol precipitation polysaccharide was composed of Gal,Glu and Man,and PI-80 was only composed of Glu and Man.All the four ethanol precipitated polysaccharides had certain antioxidant capacity.Among the three antioxidant tests,PI-80 had the best comprehensive antioxidant activity with the IC_(50) of 0.2484 mg/mL for scavenging ABTS free radicals,while PI-40 had the weakest antioxidant activity with the IC_(50) of 2.9043 mg/mL for scavenging ABTS free radicals.
