摘 要:采用分光光度法测定5种室内植物代谢甲醛的能力为吊兰>常春藤>矮牵牛>滴水观音>铁线蕨.根据GenBank中甲醛代谢途径中谷胱甘肽依赖型甲醛脱氢酶(FALDH)基因序列设计简并引物,从5种室内植物中扩增FALDH基因片段并测序.根据FALDH基因保守序列,设计荧光定量PCR引物,结果显示5种室内植物FALDH基因相对表达量与其代谢甲醛的能力一致.FALDH基因的相对表达量可作为植物代谢甲醛能力的潜在标志,研究结果可为高效准确分析植物代谢甲醛的能力提供技术支撑.The ability order of five indoor plants to metabolize formaldehyde determined by spectrophotometry was Chlorophytum comosum>Hedera nepalensis>Petunia hybrida>Alocasia macrorrhiza>Adiantum capillus-veneris.According to the gene sequence of the key enzyme formaldehyde dehydrogenase(FALDH)in the formaldehyde metabolism pathway from GenBank,degenerate primers were designed to amplify and sequence the gene fragments of FALDH from five indoor plants respectively.Based on the conserved gene sequence of FALDH,primers of FALDH were designed for fluorescent quantitative PCR.The results showed that the relative gene expression level of FALDH in five indoor plants was consistent with the formaldehyde metabolism capacity,which indicated that the gene of FALDH could be used as a molecular marker for the ability of indoor plants to metabolize formaldehyde.The research results could provide technical support for efficient and accurate analysis on the ability of plants to metabolize formaldehyde.
