摘 要:目的探讨糖磺基转移酶4(CHST4)基因对食管癌细胞增殖、侵袭和凋亡的影响。方法通过基因表达谱在线分析平台(GEPIA)分析CHST4基因在食管癌患者和正常人中的表达。通过shRNA技术干扰CHST4在食管癌细胞中的表达,然后通过实时荧光定量聚合酶链反应(RT-qPCR)检测CHST4的干扰效果。采用CCK-8、克隆形成、划痕、Transwell、流式细胞和Western blot等技术分析阴性对照组和sh-CHST4组食管癌细胞的增殖、迁移、侵袭、凋亡和相关蛋白的表达。结果通过GEPIA分析发现CHST4在食管癌患者中的表达高于正常人(P<0.05);CHST4在食管癌细胞中的表达高于正常食管上皮细胞(P<0.05);相比阴性对照组,sh-CHST4组细胞的增殖和克隆形成能力降低,细胞的迁移和侵袭能力降低,细胞凋亡增加,细胞迁移相关蛋白的表达降低(P<0.05)。结论CHST4基因在食管癌的发生和发展中起促进作用,除此之外,CHST4基因能够成为治疗食管癌的理想靶点。Objective To investigate the effect of carbohydrate sulfotransferase 4(CHST4)gene on the proliferation,invasion and apoptosis of esophageal carcinoma cells.Methods Gene expression profiling interactive analysis(GEPIA)was used to analyze the expression of CHST4 gene in patients with esophageal cancer and normal persons.The expression of CHST4 in esophageal carcinoma cells was interfered by shRNA,and the interference effect of CHST4 was detected by real time fluorescent quantitative polymerase chain reaction(RT-qPCR).The proliferation,migration,invasion,apoptosis and related protein expression of esophageal carcinoma cells in negative control group and sh-CHST4 group were analyzed by CCK-8,clone formation assay,wound-healing assay,Transwell,flow cytometry and Western blot.Results GEPIA analysis showed that the expression of CHST4 in patients with esophageal cancer was higher than that in normal persons(P<0.05).The expression of CHST4 in esophageal cancer cells was higher than that in normal esophageal apithelial cells(P<0.05).Compared with the negative control group,the sh-CHST4 group had lower cell proliferation and clonogenic ability,lower cell migration and invasion ability,higher cell apoptosis,and lower expression of cell migration-related protein(P<0.05).Conclusion CHST4 gene plays a promoting role in the development of esophageal cancer.In addition,CHST4 gene can be an ideal target for the treatment of esophageal cancer.
