前列腺癌患者癌组织中miR-647和HOXA9表达水平及其与预后的关系分析

摘　　要：目的通过检测前列腺癌组织中微小RNA(microRNA,miR)-647、同源盒A9(HOXA9)表达水平,分析miR-647、HOXA9与前列腺癌患者预后的关系。方法收集本院108例前列腺癌患者术中切除的癌组织及其对应的癌旁组织,使用qRT-PCR技术检测组织中miR-647和HOXA9 mRNA表达水平,免疫组化法检测HOXA9蛋白表达水平,并分析miR-647、HOXA9表达水平与患者临床病理特征及预后的关系;Pearson相关系数分析miR-647与HOXA9的相关性。结果前列腺癌组织中miR-647表达水平(0.65±0.12)显著低于癌旁组织(1.01±0.09)(P<0.05),HOXA9 mRNA表达水平(1.86±0.37)明显高于癌旁组织(1.03±0.10)(P<0.05);癌组织中HOXA9蛋白阳性表达率显著高于癌旁组织(P<0.05);前列腺癌组织中miR-647与HOXA9呈负相关(r=-0.680,P=0.000);miR-647、HOXA9表达水平与患者T分期、淋巴结有无转移、Gleason评分以及前列腺特异抗原(PSA)水平有关(P<0.05);生存分析表明,miR-647高表达、HOXA9低表达时患者生存率较高;miR-647、HOXA9、Gleason评分是影响患者预后的相关因素(P<0.05)。结论miR-647在前列腺癌中低表达,而HOXA9高表达,miR-647与HOXA9呈负相关,两者表达水平与部分临床病理特征以及术后3年生存率有关,可影响患者预后。Objective To investigate the correlation be tween the level of the expressions of microRNA(miR)-647 and homeobox A9(HOXA9)in prostate cancer tissues,and the clinical prognosis in the patients-prostatic.Methods The cancer tissues and their corresponding adjacent tissues of 108 prostate cancer patients who were treated in our hospital were respectively collected.The expressions of miR-647 and HOXA9 mRNA in the tissues were detected by qRT-PCR,and the expression of HOXA9 protein was analyzed by immunohistochemistry.The relationship between the expression levels of miR-647 and HOXA9 and the clinicopathological characteristics and prognosis of patients were analyzed.The correlation between miR-647 and HOXA9 was clarified using Pearson method.Results The expression level of miR-647 in prostate cancer tissues(0.65±0.12)was significantly lower than that in adjacent tissues(1.01±0.09)(P<0.05),and the expression level of HOXA9 mRNA(1.86±0.37)was significantly higher than that in adjacent tissues(1.03±0.10)(P<0.05).The positive expression rate of HOXA9 protein in cancer tissues was obviously higher than that in adjacent tissues(P<0.05).There was a negative correlation between miR-647 and HOXA9 in prostate cancer tissue(r=-0.680,P=0.000).The expression levels of miR-647 and HOXA9 were related to the T stage,lymph node metastasis,Gleason score,and prostate specific antigen(PSA)level(P<0.05).Survival analysis showed that patients with high expression of miR-647 and low expression of HOXA9 had a higher survival rate.miR-647,HOXA9 and Gleason score were related factors affecting the prognosis of patients(P<0.05).Conclusion MiR-647 is lowly expressed in prostate cancer,while HOXA9 is highly expressed.There is a negative correlation between miR-647 and HOXA9.The expression levels of both are related to some clinicopathological characteristics and the 3-year survival rate after surgery,which can affect the prognosis of patients.
