摘 要:目的探讨水通道蛋白3(AQP3)在前列腺癌中的表达及其对前列腺癌细胞迁移侵袭及上皮间质转化(EMT)的影响。方法采用实时荧光定量聚合酶链式反应(qPCR)和免疫组化(IHC)方法检测68例前列腺癌及相匹配的邻近非癌前列腺组织中AQP3的表达水平。将AQP3小干扰RNA(siRNA)转染人前列腺癌细胞(siAQP3组),同时设对照组(siNC组),qPCR和Western blotting验证转染效率。Transwell实验检测细胞迁移和侵袭能力,采用Western blot检测EMT标志物E-钙黏蛋白(E-cadherin)、N-钙黏蛋白(N-cadherin)、波动蛋白(Vimentin)、ZO-1和Snail蛋白表达。结果前列腺癌组织中AQP3表达显著高于相匹配的邻近非癌性前列腺组织(P<0.001),且与T分期、淋巴结转移和Gleason评分显著相关(P<0.05)。此外,与siNC组相比,siAQP3组细胞AQP3的mRNA及蛋白水平均降低。与siNC组比较,siAQP3组细胞增殖、迁移和侵袭能力显著降低,E-cadherin和ZO-1表达上调表达升高,Snail、N-cadherin和Vimentin表达下降(P<0.05)。结论AQP3在前列腺癌中高表达,且与不良预后有关,降低AQP3表达能逆转EMT,并降低细胞迁移和侵袭能力。Objective To investigate the expression of aquaporin 3(AQP3)in prostate cancer and its effect on the migration,invasion and epithelial-mesenchymal transition(EMT)of prostate cancer cells.Methods Real-time fluorescent quantitative polymerase chain reaction(qPCR)and immunohistochemistry(IHC)were used to detect the expressions of AQP3 in 68 prostate cancer tissues and their paired adjacent non-cancerous prostate tissues.Human prostate cancer cells transfected with AQP3 small interfering RNA(siRNA)was defined as siAQP3 group,and a control group as siNC group.The transfection efficiency was evaluated by qPCR and Western blotting.Cell migration and invasion were measured by Boyden Chamber Assay.The expressions of EMT markers such as E-cadherin,N-cadherin,Vimentin,Snail and ZO-1 were analyzed by Western blotting.Results The expression of AQP3 in prostate cancer tissues was significantly higher than that in paired adjacent non-cancerous prostate tissues(P<0.001),and its expression level was positively correlated to T stage,lymph node metastasis and Gleason score(P<0.05).Compared with those in the siNC group,the mRNA and protein levels of AQP3 in the siAQP3 group were obviously decreased(P<0.05).Compared with those of the siNC group,the cell proliferation,migration and invasion of the siAQP3 group were all significantly decreased,simultaneously,the expressions of ZO-1 and E-cadherin were increased,and the expressions of Snail,N-cadherin and Vimentin were decreased(P<0.05).Conclusion AQP3 is obviously upregulated in prostate cancer and its high expression indicates poor prognosis.Suppression of AQP3 can reverse EMT and reduce cell migration and invasion.
