circ_0072083靶向miR-142-3p调控结直肠癌细胞增殖、侵袭和迁移

摘　　要：目的探讨结直肠癌组织中circ_0072083和miR-142-3p的表达情况,分析circ_0072083靶向miR-142-3p在结直肠癌进展中的作用。方法选取2018年3月至2019年12月行手术切除的52例结直肠癌患者的结直肠癌组织和癌旁组织。RT-qPCR检测circ_0072083和miR-142-3p在结直肠癌组织和癌旁组织中的表达。将si-circ_0072083、si-NC、miR-142-3p mimic、miR-NC、si-circ_0072083+anti-miR-NC、si-circ_0072083+miR-142-3p Inhibitor分别转染结直肠癌LoVo细胞,运用平板克隆实验、Transwell法、CCK-8法评估细胞克隆、迁移侵袭和增殖能力。circ_0072083和miR-142-3p的靶向关系通过双荧光素酶报告实验确定。结果结直肠癌组织中circ_0072083表达水平显著高于癌旁组织(P<0.05),miR-142-3p表达水平显著低于癌旁组织(P<0.05)。沉默circ_0072083显著增加细胞增殖抑制率和miR-142-3p表达水平(P<0.05),减少克隆数、迁移数和侵袭数(P<0.05)。过表达miR-142-3p显著增加细胞增殖抑制率(P<0.05),减少克隆数、迁移数和侵袭数(P<0.05)。miR-142-3p是circ_0072083的直接靶点。抑制miR-142-3p表达显著减弱沉默circ_0072083对LoVo细胞增殖、迁移、克隆和侵袭的影响(P<0.05)。结论沉默circ_0072083通过靶向上调miR-142-3p能够抑制结直肠癌细胞的增殖、迁移和侵袭。Objective To explore the expressions of circ_0072083 and miR-142-3p in colorectal cancer(CRC)tissues,and to further analyze the role of circ_0072083 targeting miR 1423p on CRC progression.Methods From March 2018 to December 2019,the expressions of circ_0072083 and miR-142-3p in CRC tissues and adjacent tissues were detected by using RT-qPCR which were from 52 CRC patients receiving surgical resection.The si-circ_0072083,si-NC,miR-142-3p mimic,miR-NC,si-circ_0072083+anti-miR-NC,si-circ_0072083+miR-142-3p Inhibitor were transfected into colorectal cancer LoVo cells respectively,and the plate cloning assay,Transwell method and CCK-8 method were applied to evaluate cell cloning,migration,invasion and proliferation capacities.The targeting relation between circ_0072083 and miR-142-3p was confirmed by the dual luciferase reporter assay.Results The expression level of circ_0072083 in CRC tissue was significantly higher than that in adjacent tissues(P<0.05),and the expression level of miR-142-3p was significantly lower than that in adjacent tissues(P<0.05).Silencing circ_0072083 significantly increased the inhibition rate of cell proliferation and miR-142-3p expression level(P<0.05),and reduced the number of clones,migration and invasion(P<0.05).Over-expression of miR-142-3p significantly increased the inhibition rate of cell proliferation(P<0.05),and decreased the number of clones,migration and invasion(P<0.05).miR-142-3p was a direct target of circ_0072083.miR-142-3p inhibition significantly reduced the effects of silencing circ_0072083 on the proliferation,migration,cloning and invasion of LoVo cells(P<0.05).Conclusion Silencing circ_0072083 is capable of inhibiting the proliferation,migration and invasion of CRC cells by targeting and up-regulating miR-142-3p.
