摘  要:目的 探讨巨噬细胞移动抑制因子(MIF)基因-173G/C多态性与社区女性人乳头瘤病毒(HPV)易感性之间的关系,并分析不同基因型间MIF水平的差异。方法 收集本社区HPV感染者44例作为感染组,同期HPV阴性的健康女性46例作为对照组。采集研究对象宫颈脱落细胞检测HPV核酸,并采集外周血分离血浆检测MIF浓度;提取基因组DNA,采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)检测MIF-173G/C基因多态性。频数计数法计算各组基因型和等位基因频率,并对MIF水平进行统计学分析。结果 HPV感染组与对照组MIF基因-173位点均以GG型为主,GG型与GC/CC型(OR=1.118,95%CI:0.485~2.614)、等位基因G与C(OR=1.313,95%CI:0.636~2.628)分布频率均无差异(P>0.05)。感染组与对照组MIF浓度分别为(0.952±1.111)ng/ml、(1.304±1.230)ng/ml,两组间无统计学差异(P>0.05)。GG型MIF浓度在感染组与对照组中分别为(1.270±1.310)ng/ml、(1.479±1.335)ng/ml,差异无统计学差异(P>0.05);感染组GC/CC型MIF浓度为(0.456±0.335)ng/ml,低于对照组的(1.027±1.018)ng/ml(P=0.041)。本研究中所有研究对象,GC/CC型MIF血清浓度为(0.750±0.809)ng/ml,GG基因型MIF浓度为(1.378±1.314)ng/ml,差异具有统计学意义(P=0.008);在感染组内,GC/CC型MIF浓度为(0.456±0.335)ng/ml,同样低于GG型的(1.270±1.310)ng/ml(P=0.006)。结论 MIF-173G/C位点基因多态性可能与本地区HPV易感性无关。MIF-173位点具有C等位基因可能更易影响MIF水平,并在HPV感染及进展中发挥重要作用。Objective To explore the role of macrophage migration inhibitory factor(MIF)gene promoter-173G/C gene polymorphism in the susceptibility of HPV infection and serum MIF concentration in community women.Methods Totally 44 patients with HPV infection were assigned as the infection group,and 46 healthy women were assigned as control group.Peripheral blood with EDTA anticoagulant and clinical information of specimens were collected.The blood serum was separated for MIF testing and genomic DNA from peripheral blood of all specimens were extracted.MIF-173G/C single nucleotide polymorphisms were detected using PCR-restriction fragment length polymorphism(PCR-RFLP)method.Genotype and allele frequency were calculated using frequency counting method.The MIF concentration was detected and the difference between different groups was analyzed.Results Type GG was the predominant genotype both in infection and control group.There was no significant difference in the frequency distribution of GG,GC/CC genotype(OR=1.118,95%CI:0.485-2.614),and the allele G and C(OR=1.313,95%CI:0.636-2.628).MIF concentration of serum in infection and control group were(0.952±1.111)ng/ml and(1.304±1.230)ng/ml,respectively,with no significant difference(P>0.05).MIF concentration of serum with GG genotype in infection and control group were(1.270±1.310)ng/ml and(1.479±1.335)ng/ml,respectively,with no significant difference(P>0.05).MIF concentration of serum was(0.456±0.335)ng/ml with GG/GC genotype in infection group and(1.027±1.018)ng/ml in control group,with significant difference(P=0.041).The MIF concentration was(1.270±1.310)ng/ml with GG genotype and(0.456±0.335)ng/ml with GG/GC genotype in infection group,with significant difference (P<0.01). Compared with MIF concentration of all the samples with GC/CC genotype (0.750±0.809ng/ml), MIF concentration increased significantly in samples with GG genotype (1.378±1.314ng/ml) (P<0.01). Conclusion MIF -173G/C alleles might be irrelevant to hereditary susceptibility of HPV infection in this comm
