摘  要:目的探讨lncRNA SNHG8对膀胱癌T24细胞增殖、迁移和侵袭的影响以及其分子作用机制.方法选取本院2017年1月至2019年1月收治的30例膀胱癌患者的癌组织及相应的癌旁组织;将膀胱癌T24细胞分为si-NC组、si-SNHG8组、miR-NC组、pcDNA组、pcDNA-SNHG8组、miR-335-5p组、si-SNHG8+anti-miR-NC组及si-SNHG8+anti-miR-335-5p组.对各组细胞采用qRT-PCR、MTT、Transwell、Western blot和双荧光素酶报告实验等检测并进行比较分析.结果与癌旁组织比较,膀胱癌组织中SNHG8的表达水平升高,miR-335-5p表达水平下降(均P<0.001).抑制lncRNA SNHG8或过表达miR-335-5p后,T24细胞的活力、迁移和侵袭能力显著下降,CyclinD1、MMP-2和MMP-9蛋白表达水平降低,p21蛋白表达水平升高(均P<0.001).lncRNA SNHG8靶向调控miR-335-5p的表达水平.与si-SNHG8+anti-miR-NC组比较,si-SNHG8+anti-miR-335-5p组的细胞活性、迁移能力、侵袭细胞数、CyclinD1、MMP-2、MMP-9蛋白表达水平升高,p21蛋白表达水平降低(均P<0.001),即下调miR-335-5p表达可逆转抑制lncRNA SNHG8表达对T24细胞增殖、迁移和侵袭的影响.结论下调lncRNA SNHG8可通过促进miR-335-5p表达抑制T24细胞的增殖、迁移和侵袭.Objective To explore the effects of IncRNA SNHC8 on proliferation,migration,and invasion of bladder cancer T24 cells and its molecular mechanism.Methods Cancer tissues and matched paracancer tissues of 30 patients with bladder cancer admitted to our hospital from January 2017 to January 2019 were selected.Bladder cancer T24 cells were divided into si-NC group,si-SNHG8 group,pcDNA group,pcDNA-SNHG8 group,miR-NC group,miR-335-5p group,si-SNHC8+anti-miR-NC group and si-SNHC8+anti-miR-335-5p group.qRT-PCR,MTT,Transwell,Western blot and dual luciferase reporter assay were performed.Results Compared with paracancer tissues,SNHC8 expression level was increased and miR-335-5p expression level was decreased in bladder cancer tissues(all P<0.001).Inhibition of IncRNA SNHC8 or overexpression of miR-335-5p significantly decreased the viability,migration and invasion ability of T24 cells,decreased the protein expression levels of CyclinD1,MMP-2 and MMP-9,and increased the protein expression levels of p21(all P<0.001).IncRNA SNHG8 targets regulated the expression level of miR-335-5p.Compared with si-SNHG8+anti-miR-NC group,si-SNHG8+anti-miR-335-5p group showed increased cell activity,migration ability,number of invasive cells,protein expression levels of CyclinD1,MMP-2 and MMP-9,while decreased protein expression levels of p21(all P<0.001).Down-regulating the expression of miR-335-5p reversed the effect of IncRNA SNHG8 expression on the proliferation,migration and invasion of T24 cells.Conclusions Down regulation of lncRNA SNHC8 inhibits the proliferation,migration and invasion of T24 cells by promoting the expression of miR-335-5p.
