摘  要:目的 探讨垂体瘤转化基因(PTTG)在肾透明细胞癌中的表达及其临床意义.方法 选取2019年1月至2020年12月在本院诊断为肾透明细胞癌的69例患者为研究对象.对患者手术标本的组织切片进行PTTG染色,并收集患者的临床资料.根据PTTG在肿瘤组织中是否表达,将患者分为PTTG阳性组(43例)和PTTG阴性组(26例).通过GEPIA对癌症基因图谱(TCGA)数据库中肾透明细胞癌的基因表达数据进行分析,包括PTTG在癌和癌旁组织、不同分期肿瘤中的表达水平,并进行生存分析.结果 69例肿瘤组织中,有43例组织的PTTG表达为阳性,阳性率为62.32%,26例PTTG表达阴性,阴性率为37.68%.而69例癌旁组织中,仅11例组织可见PTTG阳性表达,阳性率为15.94%.肾透明细胞癌组织中的PTTG表达阳性率显著高于癌旁组织(χ^(2)=29.294,P<0.001).PTTG阳性组中的Fuhrman 3~4级(χ^(2)=7.057,P=0.008)、TNM Ⅲ~Ⅳ 期(χ^(2)=8.918,P=0.003)和肿瘤长径>4 cm(χ^(2)=3.907,P=0.048)的比例均显著高于PTTG阴性组.TCGA数据库分析显示,PTTG在肾透明细胞癌肿瘤组织中的表达显著高于癌旁组织(P<0.05);且PTTG在不同分期的肾透明细胞癌中表达存在显著差异,其中Ⅳ期肾透明细胞癌中PTTG的表达最高(P<0.05).此外,PTTG高表达患者的预后显著低于PTTG低表达的患者(P<0.05).结论 PTTG在肾透明细胞癌中表达增高,且与肿瘤分期、分级、长径有明显相关性,且PTTG高表达的肾透明细胞癌患者的预后较差.Objectivee To explore the expression and clinical significance of pituitary tumor transforming gene(PTTG)in renal clear cell carcinoma.Methods A total of 69 patients diagnosed with renal clear cell carcinoma in our hospital from January 2019 to December 2020 were included in this study.PTTG staining was performed on the tissue sections,and the clinical data of the patients,including gender,age,pathological stage,tissue type,etc.were collected.According to the expression of PTTG in tumor tissues,patients were divided into PTTG positive group(43 cases)and PTTG negative group(26 caases).The gene expression data of renal clear cell carcinoma in The Cancer Gene Ailas(TCGA)database were analyzed by GEPIA,including the expression level of PTTG in cancer and adjacent tissues,different stages of tumor,and survival analysis.Results Among 69 tumor tissues,PTTG expression was positive in 43 tissues(positive rate was 62.32%),and negative in 26 tissues(negative rate was 37.68%).PTTG was positively expressed in only 11 of the 69 paracancer tissues,with a positive rate of 15.94%.The positive rate of PTTG expression in renal clear cell carcinoma tumor tissue was significantly higher than that in adjacent tissue(χ^(2)=29.294,P<0.001).The proportion of Fuhrman stage 3-4(χ^(2)=7.057,P=0.008),TNM stage II-IV(χ^(2)=8.918,P=0.003)and tumor diameter>4 cm(χ^(2)=3.907,P=0.048)in PTTG-positive patients was significantly higher than that in PTTG-negative patients.TCGA database analysis showed that the expression of PTTC in renal clear cell carcinoma tumor tissue was significantly higher than that in adjacent tissue(P<0.05).The expression of PTTG was significantly different in different stages of renal clear cell carcinoma,and the expression of PTTG was highest in stage IV tumors(P<0.05).In addition,the prognosis of patients with high PTTG expression was significantly lower than that of patients with low PTTG expression(P<0.05).Conclusions The expression of PTTG in renal clear cell carcinoma is increased,and is significantly correlated w
