摘 要:以金花葵花为原料,采用响应面法优化超声提取金花葵总黄酮提取工艺。以总黄酮提取率为考察指标,在单因素试验基础上,研究乙醇浓度、浸泡时间及颗粒大小3因素对总黄酮得率的影响。通过测定金花葵花提取物的还原能力和对DPPH自由基、羟自由基及超氧阴离子的清除能力来评价金花葵花提取物的抗氧化活性。结果表明,超声提取的最佳工艺为乙醇浓度75%,浸泡时间2.1 h,颗粒大小为45目,该条件下总黄酮提取率的预测值为18.64%,实测平均值为18.03%。金花葵花提取液抗氧化活性良好,并随着提取液浓度的增加而逐渐增强。Taking Hibiseu manihot L.as raw material,response surface methodology(RSM)was used to optimize the ultrasonic extraction of total flavonoids from sunflower sunflower.Taking the extraction rate of total flavonoids as an inspection index,on the basis of single factor experiments,three factors of ethanol concentration,soaking time and particle size were selected to study the influence of each factor on the yield of total flavonoids.The antioxidant activity of the extract was evaluated by measuring its reducing ability and scavenging ability to DPPH free radicals,hydroxyl free radicals and superoxide anions.The results showed that the best ultrasonic extraction process was 75%ethanol concentration,2.1 h immersion time,and 45 mesh particle size.Under these conditions,the predicted value of total flavonoid extraction rate was 18.64%,and the measured average value was 18.03%.The antioxidant activity of the Hibiseu manihot L.extract is good,and it gradually increases with the increase of the concentration of the extract.
