乳腺癌组织中miR-9-5p、UBE3C的表达及临床意义

摘　　要：目的:分析乳腺癌组织中微小RNA-9-5p(miR-9-5p)、泛素连接酶E3C(UBE3C)的表达水平,并探讨其与预后的关系。方法:收集2015年1月—2016年1月行手术治疗的50例乳腺癌患者的临床资料及乳腺癌组织和对应的癌旁正常组织标本。采用实时荧光定量PCR法检测miR-9-5p表达水平;Ualcan数据库分析乳腺癌组织UBE3C基因表达水平;免疫组织化学法检测乳腺癌组织中UBE3C蛋白表达;分析乳腺癌组织中miR-9-5p、UBE3C表达水平与临床病理特征的关系;随访5年,分析miR-9-5p、UBE3C水平与乳腺癌患者5年累积生存率的关系。结果:检索Ualcan数据库发现乳腺癌组织中UBE3C基因表达水平高于癌旁组织(P<0.05);与癌旁组织相比,乳腺癌组织中miR-9-5p、UBE3C表达水平均升高(P<0.05);miR-9-5p、UBE3C表达水平均与淋巴结转移、TNM分期相关(P<0.05);miR-9-5p高表达患者5年累积生存率(31.80%)低于miR-9-5p低表达患者(78.60%),差异有统计学意义(χ^(2)=13.127,P<0.001);UBE3C高表达患者5年累积生存率(36.40%)低于UBE3C低表达患者(64.10%),差异有统计学意义(χ^(2)=5.636,P=0.018)。结论:miR-9-5p、UBE3C在乳腺癌组织中高表达,与乳腺癌患者部分临床病理特征有关,可影响患者预后。Objective:To analyze the expression levels of microRNA-9-5p(miR-9-5p)and ubiquitin protein ligase E3C(UBE3C)in breast cancer tissues,and to explore their relationship with the prognosis.Methods:Fifty patients with breast cancer who underwent surgical treatment from January 2015 to January 2016 were selected to collect their cancer tissues and corresponding normal tissues adjacent to the cancer.Real-time fluorescent quantitative PCR method was used to detect the expression level of miR-9-5p;Ualcan database was used to analyze the UBE3C gene expression level in breast cancer tissues;immunohistochemical method was used to detect the expression of UBE3C protein in breast cancer tissues;the relationship between the levels of miR-9-5p and UBE3C in breast cancer tissues and the clinicopathological characteristics was analyzed;the patients were followed up for 5 years,and the relationship between the levels of miR-9-5p and UBE3C and the 5-year cumulative survival rate of breast cancer patients was analyzed.Results:Searching the Ualcan database found that the UBE3C gene expression level in breast cancer tissues was higher than that in adjacent tissues(P<0.05);compared with adjacent tissues,the expression levels of miR-9-5p and UBE3C in breast cancer tissues were increased(P<0.05);the expression levels of miR-9-5p and UBE3C were correlated with lymph node metastasis and TNM staging(P<0.05);the 5-year cumulative survival rate of patients with high miR-9-5p expression(31.80%)was lower than that of patients with low miR-9-5p expression(78.60%),and the difference was statistically significant(χ2=13.127,P<0.001);the 5-year cumulative survival rate of patients with high UBE3C expression(36.40%)was lower than that of patients with low UBE3C expression(64.10%),and the difference was statistically significant(χ2=5.636,P=0.018).Conclusion:MiR-9-5p and UBE3C are highly expressed in breast cancer tissues,they are related to some clinicopathological characteristics of breast cancer patients,and can affect the prognosis of patients.
