摘　　要：目的探讨长链非编码RNA(long noncoding RNA,lncRNA)GATA结合蛋白2-AS1(lncRNA GATA2-AS1)在结直肠癌细胞系中的表达以及对结直肠癌细胞生物学功能的意义。方法通过基因表达谱交互分析(Gene Expression Profiling Interactive Analysis,GEPIA)数据库比较lncRNA GATA2-AS1在各种肿瘤中尤其是结直肠癌中的表达差异。通过实时荧光定量聚合酶链反应(real time-quantitative polymerase chain reaction,RT-qPCR)法检测结直肠癌细胞和人正常肠黏膜上皮细胞中lncRNA GATA2-AS1的表达情况。应用小干扰RNA(siRNA)沉默HCT116细胞中lncRNA GATA2-AS1的表达,通过CCK-8法、EdU增殖实验、划痕实验、Transwell侵袭实验检测lncRNA GATA2-AS1对结直肠癌细胞增殖、迁移和侵袭的影响。采用Western blot法检测上皮间充质转化相关蛋白表达情况。结果结直肠癌细胞的lncRNA GATA2-AS1表达水平与正常肠黏膜上皮细胞比较均显著升高(P<0.05)。干扰lncRNA GATA2-AS1的表达抑制了结直肠癌细胞的增殖、迁移和侵袭(P<0.05)。干扰lncRNA GATA2-AS1后,E-cadherin蛋白上调,N-cadherin蛋白明显下调。结论lncRNA GATA2-AS1在结直肠癌中表达显著增加,且与结直肠癌的增殖、迁移、侵袭及上皮间充质转化相关。Objective To investigate the expression of long noncoding RNA GATA2-AS1 in colorectal cell lines,and explore its significance in the biological function of colorectal cancer cells.Methods The expression difference of lncRNA GATA2-AS1 in various cancer samples were analyzed via Gene Expression Profiling Interactive Analysis(GEPIA)database,expecially in the colorectal cancer.The expression levels of lncRNA GATA2-AS1 in colorectal cancer cells and human normal intestinal mucosal epithelial cells were detected by real time-quantitative polymerase chain reaction(RT-qPCR).siRNA was used to silence the expression of lncRNA GATA2-AS1 in HCT116 cells.The functions of lncRNA GATA2-AS1 on proliferation,invasion and migration were analyzed by CCK-8 assay,EdU proliferation assay,Transwell assays,The expression of epithelial-mesenchymal transition related proteins was detected by Western blot.Results The expressions level of lncRNA GATA2-AS1 were significantly higher in colorectal cancer cells than that in human normal intestinal mucosal epithelial cells(P<0.05).The proliferation,migration and invasion ability of colorectal cancer cells were inhibited by interfering the expression of lncRNA GATA2-AS1(P<0.05).After interfered with lncRNA GATA2-AS1,E-cadherin protein was up-regulated and N-cadherin protein was significantly down-regulated.Conclusion The expression of lncRNA GATA2-AS1 was upregulated in colorectal cancer,and it is associated with the proliferation,migration,invasion and epithelial-mesenchymal transition of colorectal cancer.
