摘 要:目的观察柴术理胃饮对功能性消化不良(functional dyspepsia,FD)大鼠胃肠运动、下丘脑组织及p-P38MAPK通路的影响,探讨该方治疗FD的作用机制。方法SPF级健康雄性SD大鼠60只,应用慢性疲劳、悬尾刺激和饮食失节方法建立FD模型,按随机数字表法将其分为空白组,模型组,柴术理胃饮低、中、高剂量组和多潘立酮组,每组10只,分别给予相应药物治疗,观察大鼠的一般状态,记录进食量和体质量,观察胃肠功能变化、下丘脑组织病理学改变,Western blot法和RT-PCR法检测大鼠胃窦组织中p-P38MAPK蛋白和基因表达。结果各给药组大鼠治疗后的进食量和体质量、胃排空率和小肠推进率较模型组改善,差异均有统计学意义(P<0.05);不同剂量的柴术理胃饮均能不同程度地降低大鼠胃窦组织中p-P38MAPK蛋白和基因表达,差异均有统计学意义(P<0.05)。结论柴术理胃饮通过改善FD大鼠的胃肠运动和降低大鼠胃窦组织中p-P38MAPK蛋白和基因表达来治疗FD。Objective To observe the effects of Chaizhu Liwei Yin on gastrointestinal motility,hypothalamus and p-P38MAPK pathway in functional dyspepsia(FD)rats,and to explore the action mechanism of this formula in the treatment of FD.Methods A total of 60healthy male SD rats of SPF grade were used to establish the FD model by chronic fatigue,tail suspension stimulation and diet abstinence.They were randomly divided into the blank group,the model group,the low,medium and high dose groups of Chaizhu Liwei Yin and domperidone group according to the random number table method,with 10 rats in each group.The rats were treated with corresponding drugs,and the general state was observed.The food intake and weight were recorded.The changes of gastrointestinal function and pathological changes of hypothalamus were observed.Western blot and RT-PCR were used to detect the expression of p-P38MAPK protein and gene in rat gastric antrum.Results After treatment,the food intake,body weight,gastric emptying rate and intestinal propulsion rate in each administration group were improved compared with those in model group,the difference were statistically significant(P<0.05);the protein and gene expressions of p-P38MAPK in gastric antrum tissue of rats were decreased by different doses of Chaizhu Liwei Yin,the difference were statistically significant(P<0.05).Conclusion Chaizhu Liwei Yin can treat FD by improving gastrointestinal motility and reducing the expression of p-P38MAPK protein and gene in gastric antrum tissue of FD rats.
