摘  要:目的 基于生物信息学挖掘结肠癌预后相关生物标志物。方法 利用基因表达汇编数据库下载结肠癌芯片数据集GSE25070和GSE115313,获取结肠癌组织样本43例、正常结肠组织样本44例,筛选出两个芯片数据集的共同差异基因。构建共同差异基因的蛋白互作网络,采用基因本体论、京都基因和基因组百科全书分析功能和通路。从癌症基因组图谱数据库获取结肠癌组织样本454例,利用R软件对共同差异基因表达水平与结肠癌患者预后的关系作整体分析,筛选出可能的结肠癌预后相关生物标志物。绘制Kaplan-Meier生存曲线比较各个基因高、低表达组患者生存率,采用单因素Cox回归分析结肠癌患者预后的影响因素,以验证结肠癌预后相关生物标志物。结果 筛选得到共同差异基因19个,包括表达上调基因2个、表达下调基因17个;主要存在于肾素分泌和胰腺分泌通路;涉及的生物过程有蛋白质三聚体,细胞成分有细胞外泌体、质膜的整合成分和膜的锚定成分等,分子功能有细胞内钙激活氯离子通道活性、氯离子通道活性。CLCA1、ITLN1和PARM1是结肠癌预后相关生物标志物。结论 CLCA1、ITLN1和PARM1是结肠癌预后相关生物标志物。Objective To screen the biomarkers significantly related to the prognosis of patients with colon cancer through bioinformatics data mining.Methods The colon cancer microarray data sets GSE25070 and GSE115313 were downloaded using the gene expression compilation database,43 colon cancer tissue samples and 44 normal colon tissue samples were obtained,and the common differential genes of the two microarray data sets were screened out.A protein interaction network of common differential genes was constructed,and Gene Ontology,Kyoto Gene and Genome databases were used to analyze functions and pathways.A total of 454 colon cancer tissue samples were obtained from the Cancer Genome Atlas database.R software was used to conduct an overall analysis of the relationship between the expression levels of common differential genes and the prognosis of colon cancer patients,and possible biomarkers related to colon cancer prognosis were screened.Kaplan-Meier survival curves were drawn to compare the survival rates of patients in the high and low expression of each gene,and the factors influencing the prognosis of colon cancer patients were analyzed by univariate Cox regression.Results A total of 19 differentially expressed genes were screened out,including 2 up-regulated genes and 17 down-regulated genes.They were mainly related to renin secreting and pancreatic secreting pathways,involving in biological processes such as protein homotrimerization,cellular components such as extracellular exosome,integral component of plasma membrane and anchored component of membrane,etc.The molecular functions included intracellular calcium activated chloride channel activity and chloride channel activity.Conclusion The study shows that CLCA1,ITLN1and PARM1 might be the prognostic biomarkers in colon cancer.
