摘  要:为了筛选得到高产γ-氨基丁酸(Gamma-aminobutyric acid,GABA)菌株,本试验以从云南不同地区食品中分离出的27株菌为材料,通过菌株复壮、革兰氏染色形态学观察、过氧化氢酶实验、单菌株发酵及高效液相色谱法检测GABA,筛选其中高产菌株进行种属鉴定并测定其发酵性能。结果表明,27株菌中筛选得到的LSR-L-L4为高产GABA菌株,其发酵液中GABA检测值为490.59 mg/L;经16S rDNA基因序列分析鉴定其为德氏乳杆菌保加利亚亚种(Lactobacillus delbrueckii subsp.Bulgaricus),最适发酵温度为42℃;3~12代菌株凝乳时间4.5~5h,酸度52~56°T,120h极限酸度118~122°T;生长曲线稳定期OD600=1.01~1.11,24h未见衰退;活菌数1.20×10^(9)~1.50×10^(9)cfu/mL;双乙酰含量10.32~11.50 mg/L;γ-氨基丁酸产值467.58~490.59 mg/L。LSRL-L4高产γ-氨基丁酸,兼具优质发酵特性,为后续研发含γ-氨基丁酸乳制品奠定了工业基础。In order to screen strains with high production of gamma-aminobutyric acid(GABA),27 strains isolated from food in different areas of Yunnan were used as materials in this experiment.Some efforts was performed by strain rejuvenating,gram staining morphological observation,catalase experiment,single strain fermentation and high performance liquid chromatography to detect the content of GABA to screen the high-yielding strains.Besides,species identification and fermentation performance determination were also conducted.The results showed that LSR-L-L4 was a strain with high GABA production from 27 strains,and the detected GABA value in its fermentation broth was 490.59mg/L.It was identified as Lactobacillus delbrueckii subsp.Bulgaricus by 16S rDNA sequence analysis.The optimum fermentation temperature was 42℃.The milk coagulating time of 3-12 strains was 4.5-5h,the acidity was 52-56°T,and the limit acidity was 118-122°T at 120h.OD600=1.01-1.11 in the stable period of growth curve,no decline was observed in 24h.Viable count was 1.20-1.50×10^(9)cfu/mL.Diacetyl content was 10.32-11.50mg/L;the output value ofγ-aminobutyric acid is 467.58-490.59mg/L.LSR-L-L4 has high production ofγ-aminobutyric acid and high quality fermentation characteristics,which lays an industrial foundation for the subsequent research and development ofγ-aminobutyric acid dairy products.
