摘 要:γ-分泌酶复合物是许多1型跨膜蛋白的关键水解酶。对于Notch受体的激活和靶基因转录的调节非常重要。Notch通路的异常激活和表达与包括乳腺癌等的许多肿瘤类型的发生和发展密切相关。本综述介绍了乳腺癌的治疗现状及研究进展、γ-分泌酶复合物的基本情况、γ-分泌酶在Notch通路中的生物学功能和作用、过去5年中γ-分泌酶抑制剂在乳腺癌治疗中的进展、γ-分泌酶介导的乳腺癌的作用机制以及γ-分泌酶抑制剂的优缺点,提出了γ-分泌酶抑制剂进一步研究和发展的前景。γ-secretase complex is the key hydrolase of type 1 transmembrane protein.It is very important for Notch receptor activation and target gene transcription regulation.The abnormal activation and expression of Notch pathway are closely related to the occurrence and development of many tumor types,including breast cancer.This review summarizes the treatment status and research progress of breast cancer,the basic situation of γ-secretase complex,the biological function and role of γ-secretase in Notch pathway,the progress of γ-secretase inhibitors in the treatment of breast cancer in the past five years,the mechanism of γ-secretase mediated breast cancer and the advantages and disadvantages of γ-secretase inhibitors.The prospect of further research and development of γ-secretase inhibitors is proposed.
