摘　　要：目的:检测卵巢浆液性癌中骨膜蛋白(POSTN)的表达特征,分析其对细胞增殖和凋亡的调控作用。方法:选择41例卵巢浆液性癌患者作为研究对象,选择41例卵巢浆液性囊腺瘤患者作为对照,均留取术后病变组织。应用免疫组化法检测POSTN、增殖细胞核抗原(PCNA)和Bcl-2相关X蛋白(BAX)的表达。选择人卵巢癌细胞株SK-OV-3和OVCAR-3,选择人正常卵巢上皮细胞株IOSE80,基于SK-OV-3和OVCAR-3细胞株转染小干扰RNA-POSTN分别建立si-POSTN组,并设立空白对照组,应用Western blot检测POSTN、PCNA和BAX的表达,应用CCK-8法检测细胞增殖活性,应用流式细胞术观察细胞凋亡。结果:卵巢浆液性癌组织中POSTN、PCNA的阳性率明显高于卵巢浆液性囊腺瘤组织,卵巢浆液性癌组织中BAX的阳性率明显低于卵巢浆液性囊腺瘤组织(均P<0.05)。相关分析显示卵巢浆液性癌组织中POSTN和PCNA呈正相关性(r=0.68,P=0.0241)、POSTN和BAX呈负相关性(r=-0.62,P=0.0348)。SK-OV-3和OVCAR-3细胞株中POSTN的表达均明显高于JOSE80(均P<0.05)。相对于SK-OV-3和OVCAR-3细胞株的空白对照组,si-POSTN组的POSTN、PCNA的表达均下降,BAX的表达均升高,si-POSTN组均在72 h时细胞增殖活性明显下降,并持续至96 h,且细胞凋亡率均升高(均P<0.05)。结论:POSTN在卵巢浆液性癌中高表达,可能通过促进肿瘤细胞增殖、抑制凋亡发挥作用。Objective:To detect the expression characteristics of Periostin(POSTN)in ovarian serous carcinoma,and to analyze its regulatory effects on cell proliferation and apoptosis.Methods:A total of 41 patients with ovarian serous carcinoma were selected as the study group,and 41 patients with ovarian serous cystadenoma were selected as the control group.The postoperative pathological tissues were collected.The expressions of POSTN,proliferating cell nuclear antigen(PCNA)and Bcl-2 associated X protein(BAX)were detected by immunohistochemistry.Human ovarian cancer cell lines SK-OV-3 and OVCAR-3 and human normal ovarian epithelial cell line IOSE80 were selected.The human ovarian cancer cell lines SK-OV-3 and OVCAR-3 were selected,and the human normal ovarian epithelial cell line IOSE80 was selected,and the si-POSTN group was established based on the transfection of SK-OV-3 and OVCAR-3 cell lines with small interfering RNA-POSTN,and a blank control group was established.Western blot was used to detect the expression of POSTN,PCNA and BAX.CCK-8 method was used to detect the cell proliferation activity,and flow cytometry was used to observe the cell apoptosis.Results:The positive rates of POSTN and PCNA were significantly higher in ovarian serous carcinoma than those in ovarian plasmacytoma tissues,and the positive rate of BAX was significantly lower in ovarian serous carcinoma than that in ovarian plasmacytoma tissues(all P<0.05).Positive correlation was found between POSTN and PCNA(r=0.68,P=0.0241),negative correlation was found between POSTN and BAX(r=-0.62,P=0.0348)in ovarian serous carcinoma.The expression of POSTN was significantly higher in SK-OV-3 and OVCAR-3 cell lines than that in JOSE80(all P<0.05).Relative to the blank control group of SK-OV-3 and OVCAR-3 cell lines,the expression of POSTN and PCNA decreased and the expression of BAX increased,cell proliferative activity decreased at 72 hours and continued until 96 hours,apoptosis rate increased in the si-POSTN group(all P<0.05).Conclusion:POSTN is highly expresse
