摘  要:在水温(28.0±1.0)℃下,将斑马鱼(Danio rerio)受精卵暴露于2-乙基己酯-4-甲氧基肉桂酸(2-ethylhexyl-4-methoxycinnamic acid,EHMC)中,浓度为0μg·L^(-1)(对照组,仅0.1%DMSO助溶剂)、2μg·L^(-1)、20μg·L^(-1)和200μg·L^(-1),每个浓度三个平行。每24 h更换四分之一相应浓度的EHMC,48 h、72 h、96 h后观察死亡和畸形,测量鱼心率、体长等,120 h后测定幼鱼的生长激素(growth hormone,GH)和胰岛素样生长因子(insulin-like growth factor,IGF-Ⅰ)含量及GH/IGF-I轴相关基因的m RNA表达和转录水平。结果表明:EHMC暴露未显著影响斑马鱼的孵化率(P>0.05),但使心率显著下降(P<0.05);高浓度EHMC(200μg·L^(-1))下斑马鱼畸形率升高,幼鱼体长下降(P<0.05)。EHMC诱导的生长抑制效应可能与鱼体内IGF-I浓度下降及ghra、igf-1等基因的下调表达有关。本研究结果可为评价EHMC的水生生态安全性提供参考依据。The fertilized eggs of zebrafish(Danio rerio)were exposed to 2-ethylhexyl-4-methoxycinnamic acid(EHMC)at concentrations of 0 μg·L^(-1)(control group, only 0.1% DMSO cosolvent), 2 μg·L^(-1), 20 μg·L^(-1), and 200 μg·L^(-1), with three parallels for each concentration at a water temperature of(28.0±1.0)℃, 1/4 of the corresponding concentration of EHMC was replaced every 24 h.Death and deformity rates were observed and the heart rate and body length of the fish were measured in 48 h, 72 h, and 96 h. After120 h, the levels of growth hormone(GH)and insulin-like growth factor(IGF-I)and the m RNA expression and transcription levels of GH/IGF-I axis-related genes were detected in juvenile fish. The results showed that there were significantly lower body length and higher heart rate and malformation in the zebrafish exposed to high concentration of EHMC(200 μg·L^(-1))(P < 0.05), without significant effect on the hatching rate(P > 0.05). EHMC-induced growth inhibition may be attributed to the decrease in IGF-I concentration and the down-regulation of ghra and igf-1 m RNA levels. The findings provide a reference for the aquatic ecological safety assessment of EHMC.
