摘  要:目的:探讨水苏碱对卵巢癌细胞的抑制作用及其对内质网应激(ERS)介导的内源性凋亡通路的影响。方法:体外实验比较空白对照组(Con)与水苏碱低、中、高剂量组(1、8、16μmol/L)卵巢癌细胞存活率、凋亡率、凋亡基因mRNA和蛋白表达差异。裸鼠移植瘤模型检测水苏碱对卵巢癌细胞体内成瘤的影响。结果:与Con组比较,水苏碱低、中、高剂量组卵巢癌SKOV3细胞增殖明显受抑制,凋亡率明显升高,Bcl-2 mRNA表达明显降低,Caspase-3 mRNA及CHOP、GRP78蛋白表达明显升高,且具有剂量依赖性(均P<0.05)。结论:水苏碱可抑制卵巢癌SKOV3细胞增殖并促进其凋亡,其作用机制可能为激活ERS介导的内源性凋亡通路。Objective:To investigate inhibitory effect of stachydrine on apoptosis of ovarian cancer cell and its regulatory mechanism on endoplasmic reticulum stress(ERS)mediated endogenous apoptosis pathway.Methods:In vitro experiments,differences on survival rate,apoptosis rate,mRNA and protein expressions of apoptotic genes between blank control group(Con)and stachydrine low,medium and high doses groups(1,8,16μmol/L)were compared.Nude mice transplanted tumor model was used to test effect of stachydrine on tumor formation of ovarian cancer cells in vivo.Results:Compared with Con group,proliferations of ovarian cancer cells SKOV3 in stachydrine low,medium,and high doses groups were significantly inhibited,apoptosis rates were significantly increased,mRNA expression of Bcl-2 was decreased significantly,mRNA expression of Caspase-3,protein expressions of CHOP and GRP78 were increased significantly,which in a dose-dependent manner(all P<0.05).Conclusion:Stachytine can inhibit proliferation of ovarian cancer cells SKOV3 and promote its apoptosis,whose mechanism may be activation of ERS mediated endogenous apoptosis pathway.
