摘　　要：肝细胞功能缺失导致的肝衰竭,危及患者生命,肝移植是其唯一有效的治疗方式。然而供肝资源极其有限,肝衰竭患者常因等不到肝源而错过治疗时机乃至失去生命。肝样细胞是一类体外获得的具有肝细胞类似表达谱和类似功能的细胞,有望用于移植治疗以及药物筛选。随着对肝细胞发育的认识日渐加深,以及多能干细胞的研究发现,已经为生产用于移植疗法和药物筛选的肝样细胞提供了前所未有的机会。另外,利用已确定为正常发育过程所需的关键转录因子,在体内和体外诱导条件下使肝细胞恢复增殖能力,以及将其他终末分化的细胞类型转分化为肝样细胞方面的研究也取得了进展。在此,本文总结了各类细胞来源及其生成肝样细胞的研究策略,如何对肝样细胞进行大规模生产以及肝样细胞用于治疗的各种方式方法。A large number of hepatocytes,which loss of corresponding functions,will lead to liver failure,which is life-threatening for patients.Liver transplantation is the only effective treatment for this disease.However,the resources of donor livers are extremely scarce,and patients with liver failure often miss the opportunity for treatment or even lose their lives because they cannot wait for a liver resource for transplantation.Hepatocyte-like cells are a class of cells obtained in vitro with similar expression profiles and functions of hepatocytes,which are expected to be used for cell transplantation therapy of end-stage liver disease,or drug screening of liver-related diseases.The growing understanding of hepatocytes development and the discovery of pluripotent stem cells have provided unprecedented opportunities to produce hepatocyte-like cells for transplantation therapy and drug screening.In addition,using key transcription factors identified as necessary for normal liver development,researchers can restore hepatocytes to proliferative capacity under induced conditions in vivo and in vitro,or transdifferentiate other terminally differentiated cell types into hepatocyte-like cells.This paper summarize various cell sources and research strategies for generating hepatocyte-like cells,how to mass-produce hepatocyte-like cells,and various ways and methods for hepatocyte-like cells to be used in therapy.
