摘 要:目的:探讨雄激素受体(AR)和黑素亲和素(MLPH)在乳腺癌中的表达情况,并分析其相关性。方法:通过公共数据库中AR与MLPH的基因表达情况,分析其相关性及预后。免疫组织化学法检测62例非特殊浸润性乳腺癌患者癌组织和癌旁组织中AR和MLPH的蛋白表达,分析AR、MLPH表达与临床病理参数的关系。免疫组织荧光法检测AR与MLPH在乳腺癌组织中的表达定位情况。结果:数据库显示,AR和MLPH均在乳腺癌中高表达,表达呈正相关(r=0.73,P<0.05)。AR和MLPH高表达患者的生存期(OS)、无复发生存期(RFS)和无远处转移生存期(DMFS)均优于低表达患者(P<0.05)。免疫组化结果提示,AR表达水平与组织学分级、ER、PR、HER-2、Ki-67和远处转移有关(P<0.05),与年龄、TNM分期、肿瘤大小和绝经状态无关(P>0.05);MLPH表达水平与组织学分级、ER、PR、HER-2、Ki-67和远处转移有关(P<0.05);与年龄、TNM分期、肿瘤大小和绝经状态无关(P>0.05)。Spearman相关性分析显示,AR与MLPH的表达在乳腺癌中呈正相关(r=0.471,P<0.001)。进一步比较AR和MLPH共表达与组织学分级、ER、PR、Ki-67及远处转移的关系,阳性组与ER、PR和HER-2呈正相关(P<0.05),与组织学分级、Ki-67及远处转移呈负相关(P<0.05)。免疫荧光结果表明,AR除在细胞核上高表达,还与MLPH共表达于细胞质中,两者的表达存在共定位。结论:乳腺癌中AR和MLPH高表达,二者呈正相关,可能协同作用于乳腺癌的发生发展,为临床预后相关的潜在生物靶标。Objective:To investigate the expression of androgen receptor(AR)and melanophilin(MLPH)in breast cancer and analyze their correlation.Methods:The gene expression of AR and MLPH was detected in pubilc database,and to analyze the correlation and prognosis.Immunohistochemical method was used to detect the expression of AR and MLPH protein in cancer tissues and adjacent tissues of 62 patients of non-special invasive breast cancer.The relationship between AR and MLPH expression and clinicopathological parameters was analyzed.Immunohistofluorescence was used to detect the expression and localization of AR and MLPH in breast cancer tissues.Results:The database showed that AR and MLPH were highly expressed in breast cancer,and the expressions were positively correlated(r=0.73,P<0.05).The overall survival(OS),recurence-free survival(RFS)and distant metastasis-free survival(DMFS)of patients with high AR and MLPH expression were better than those with low expression(P<0.05).Immunohistochemical results showed that the expression level of AR was related to histological grade,ER,PR,HER-2,Ki-67 and distant metastasis(P<0.05),there was not related to age,TNM stage,tumor size and menopausal status(P>0.05).The expression level of MLPH was related to histological grade,ER,PR,HER-2,Ki-67 and distant metastasis(P<0.05),there was not related to age,TNM stage,tumor size and menopausal status(P>0.05).Spearman correlation analysis showed that there was a positively correlation between AR and MLPH expression(Rs=0.471,P<0.001).The relationship between AR and MLPH co-expression and histological grade,ER,PR,Ki-67 and distant metastasis was further compared.The positive group was positively correlated with ER,PR and HER-2(P<0.05),and negatively correlated with histological grade,Ki-67 and distant metastasis(P<0.05).Immunofluorescence results showed that AR was highly expressed in the nucleus and co-expressed with MLPH in the cytoplasm,and there was colocalization between the two expressions.Conclusion:The high expression of AR and MLPH is posi
