摘 要:利用1株由本实验室分离保存的牦牛源产气荚膜梭菌Qinghai-1菌株,首先通过生化试验、药敏试验、PCR技术对该菌株生物学特性进行了研究,随后通过全基因组测序利用生物基因数据库对基因组的基因功能进行注释和耐药基因分析。生物学特性显示,该菌株为1株牦牛源A型产气荚膜梭菌,菌株在硫化氢、乳糖、葡萄糖、蔗糖、麦芽糖、赖氨酸、鸟氨酸、卵磷脂酶、明胶、硝酸盐、甘露糖、酪蛋白共12种生化鉴定管中成阳性反应;对头孢噻肟、氨苄西林、青霉素、四环素敏感,对庆大霉素、复方新诺明、多黏菌素B、链霉素、杆菌肽耐药。全基因组分析显示,该菌株基因组长度为3042374 bp,GC含量为28.58%,共含2906个基因,2721个CDS区,94段tRNA,10段rRNA,60段miscRNA,1段tmRNA,968段重复序列和1个基因岛,不含CRISPR序列。在GO、KEGG、COG数据库中分别有1643,849,1196个基因功能注释,在VFDB数据库中有237个与毒力因子相关的基因,在CARD数据库中仅有Clostridium perfringens mprF一个抗性基因注释。提交基因组序列至NCBI网站,获得登录号:CP092481。本研究完成了对Qinghai-1菌株的耐药特性研究和基因组信息分析,丰富了产气荚膜梭菌基因库宿主的类型,为进一步了解牦牛源产气荚膜梭菌遗传进化信息、基因功能、耐药特性提供了理论参考。The biological characteristics of a yak-derived Clostridium perfringens(C.perfringens)Qinghai-1 strain isolated and preserved in our laboratory were studied by biochemical tests,drug susceptibility tests and PCR techniques,then the whole genome of the strain was sequenced and its gene function was annotated,its drug resistance genes were analyzed using the biological gene database.The biological characteristics showed that the strain was a yak-derived C.perfringens type A,and the strain showed positive reaction in 12 biochemical identification tubes including hydrogen sulfide,lactose,glucose,sucrose,maltose,lysine,ornithine,lecithinase,gelatin,nitrate,mannose,casein;the strain was sensitive to cefotaxime,ampicillin,penicillin and tetracycline,it was resistant to gentamicin,cotrimoxazole,polymyxin B,streptomycin and bacitracin.Whole genome analysis showed that the genome of this strain was 3042374bp in length,with a GC content of 28.58%,containing 2906genes,2721CDS regions,94tRNAs,10rRNAs,60miscRNAs,one tmRNA,968repeat sequences and one gene island,without CRISPR sequences.There were 1643,849,and 1196gene function annotations in the GO,KEGG,and COG databases,respectively,237genes related to virulence factors in the VFDB database,and only one resistance gene annotation,C.perfringens mprF,in the CARD database.The genome sequence was submitted to the NCBI website and the accession number was obtained:CP092481.This study completed the research on the drug resistance characteristics of Qinghai-1strain and the analysis of genomic information,enriched the types of C.perfringens gene bank hosts,and provided a theoretical reference for further understanding the genetic evolution information,drug resistance characteristics and gene function of yak-derived C.perfringens.
