摘 要:目的 探讨壬基酚(NP)暴露与维生素D_(3)(VD_(3))及钙离子(Ca^(2+))水平的量效关系,并明确NP对VD_(3)与Ca^(2+)代谢的影响。方法 通过慢性染毒实验,在无特定病原体(SPF)级BALB/C裸鼠分别暴露于低、中、高3种浓度的NP(分别为低、中、高剂量组) 60 d,选取未经染毒(给予等体积玉米油)的裸鼠作为对照,分别采用ELISA与分光光度法对血清中VD_(3)和Ca^(2+)水平变化情况进行检测,采用SPSS23.0软件对所得数据进行统计分析。结果 随着NP灌胃水平的增加,BALB/C裸鼠血清中VD_(3)水平逐渐升高,高剂量组VD_(3)水平最高,为(116.96±6.67)pg/mL,明显高于对照组的(80.11±7.29)pg/mL(P<0.05);BALB/C裸鼠血清中Ca^(2+)水平随NP灌胃水平的增加先升高后降低,其中中剂量组Ca^(2+)的水平最高,为(42.86±2.22)μmol/dL,明显高于对照组的(31.58±2.11)μmol/dL(P<0.05),而高剂量组最低,为(24.16±2.34)μmol/dL,明显低于对照组的(P<0.05)。结论 低、中剂量NP在调控VD_(3)方面发挥拟雌激素作用,当继续增加剂量时这种拟雌激素的作用被其本身的毒性所抵消。Objective To explore the dose effect relationship between nonylphenol(NP)exposure and serum vitamin D 3(VD 3)and Ca^(2+)levels,in order to preliminarily clarify the effect of NP on VD_(3) and Ca^(2+)metabolism.Methods SPF BALB/C nude mice were exposed to low,medium and high concentrations(the low,medium and high dose groups)of NP for 60 days,and the nude mice without exposure(given equal volume of cornoil)were selected as the control group,the changes of VD 3 and Ca^(2+)in serum were detected by ELISA and spectrophotometry,and the data were statistically analyzed by SPSS 23.0 software.Results With the increase of NP concentration by gavage,the level of VD 3 in the serum of mice gradually increased(low<medium<high).The level of VD_(3) in the high-dose group was the highest[(116.96±6.67)pg/mL]which was significantly higher than that in the control group[(80.11±7.29)pg/mL];The level of Ca^(2+)in serum of mice changed dramatically,that is,from high to low.The concentration of Ca^(2+)in the medium dose NP group was the highest[(42.86±2.22)μmol/dL],which was significantly higher than that in the control group(31.58±2.11)μmol/dL,while the concentration in the high dose group was the lowest[(24.16±2.34)μmol/dL],which was significantly lower than that in the control group.Conclusion Low and medium doses of NP play an estrogen like role in regulating VD_(3).When the dose continues to increase,this estrogen like effect is offset by its own toxicity.
