摘 要:运用文献计量学方法,从基因挖掘的角度探究阿尔茨海默症早期诊断的研究进展。以国内外AD基因研究的相关文献为研究对象,将CNKI和WOS核心集数据库作为文献来源,从文献计量学视角对主要研究机构、研究主题等数据信息进行外部特征分析,基于知识图谱进行内部语义挖掘以探求该领域的研究脉络和发展方向。研究发现,国内外的研究热点集中于AD的诊治方面,主要体现在潜在生物标志物的发掘和基因体细胞的识别、与AD有关的其他退行性疾病的异病同症基因表达机制研究、探索丰富基因治疗手段等三大方面。基因芯片筛选技术(如miRNAs表达改变)、CRISPR/Cas9介导的基因编辑技术、基因学检测技术(如尝试长序列三代测序)等新技术的研发与应用将成为未来研究重点。This study explores the research progress of early diagnosis of Alzheimer's disease(AD)from the perspective of gene mining through the bibliometrics.The research object is related literatures on AD gene at home and abroad,and the sources are CNKI and WOS core set databases.From the perspective of bibliometrics,the external characteristics of data information such as major research institutions and research topics are analyzed,and the internal semantic mining is carried out based on the mapping knowledge domains,so as to explore the research context and development direction of this field.It was found that research hotspots at home and abroad were focused on the diagnosis and treatment of AD,mainly in three major areas:discovery of potential biomarkers and identification of gene somatic cells,research on the expression mechanism of allopathic genes in other degenerative diseases related to AD,and exploration of enriched gene therapy tools.The development and application of new technologies such as gene microarray screening technologies(e.g.altered expression of miRNAs),CRISPR/Cas9-mediated gene editing technologies,and genetic testing technologies(e.g.attempting long sequence triple sequencing)will be the focus of future research.
