摘  要:从头测序技术是应用于蛋白质组学的测序方法。与其他测序技术相比,从头测序技术的优势是在不依赖数据库搜索和无需PCR扩增的基础上对DNA或者蛋白质进行测序。目前,从头测序技术也广泛应用于蛋白质组学领域。研究人员为了将测序技术所得的短序列reads进一步组装成一条完整的序列,就需要使用序列组装方法。首先,介绍了测序技术的发展背景、从头测序技术的概念以及序列组装算法的出现;其次,总结了基于Overlap⁃Layout⁃Consensus策略、deBruijn图策略和Greedy策略的序列组装算法的具体概念和成果;最后,阐述了序列组装算法面临的挑战。de novo sequencing technology is a sequencing method applied to proteomics.The advantage of de novo sequencing over other sequencing technologies is that DNA or protein can be sequenced without database search or PCR amplification.At present,de novo sequencing technology is also widely used in proteomics.In order to further assemble short sequence reads generated by sequencing technology into a complete sequence,researchers need to use sequence assembly methods.Firstly,the development background of sequencing technology,the concept of de novo sequencing technology and the emergence of sequence assembly algorithms are introduced;secondly,the specific concepts and results of sequence assembly algorithms based on Overlap-Layout-Consensus strategy,de Bruijn Graph strategy and Greedy strategy are summarized;finally,the challenges facing the sequence assembly algorithm are described.
