苦参碱诱导人肺腺癌细胞A549凋亡并抑制迁移的初步探讨

摘  要:目的:探讨不同浓度的苦参碱对人肺腺癌细胞A549凋亡及迁移能力的影响及分子机制。方法:以含有0.2、0.8、1.2、2.4、4.8 mg/mL浓度梯度苦参碱的培养基培养的A549细胞作为实验组,不含药物培养基培养的细胞作为对照组。细胞划痕试验检测各组A549细胞的迁移能力;RT-qPCR检测各组细胞中凋亡相关基因Bcl-2、Survivin、Caspase-9的表达;Western blotting检测各组细胞中凋亡相关蛋白Bcl-2、Survivin、Caspase-9的表达。结果:与对照组相比,1.2、2.4、4.8 mg/mL药物浓度组细胞迁移面积及迁移率减少,差异有统计学意义(P<0.05);RT-qPCR和Western blotting结果显示,苦参碱能下调凋亡相关抑制基因及蛋白Bcl-2、Survivin的表达,上调促凋亡相关基因及蛋白Caspase-9的表达,差异有统计学意义(P<0.05)。结论:苦参碱能够有效地抑制肺腺癌A549细胞体外迁移能力,并提高Caspase-9基因和蛋白表达,抑制Bcl-2、Survivin基因和蛋白表达,促进A549细胞凋亡。Objective To investigate the effects of different concentrations of matrine on the apoptosis and migration ability of human lung adenocarcinoma cells A549 and the molecular mechanism.Methods A549 cells cultured in medium containing 0.2,0.8,1.2,2.4 and 4.8 mg/mL gradients of matrine were used as the experimental group,and cells cultured in medium without the drug were used as the control group.The expression of apoptosis-related genes Bcl-2,Survivin and Caspase-9 in each group was detected by RT-qPCR and the expression of apoptosis-related proteins Bcl-2,Survivin and Caspase-9 in each group was detected by western blotting.Results Compared with the control group,the expression of apoptosis-related inhibitory genes and proteins Bcl-2 and Survivin were down-regulated and the expression of pro-apoptosis-related genes and proteins Caspase-9 were up-regulated by RT-qPCR and Western blotting(P<0.05).The results of western blotting showed that matrine could down-regulate the expression of apoptosis-related inhibitory genes and proteins Bcl-2 and Survivin,and up-regulate the expression of pro-apoptosis-related genes and protein Caspase-9(P<0.05).Conclusion Matrine can effectively inhibit the migration ability of lung adenocarcinoma A549 cells in vitro,and increase the expression of Caspase-9 genes and proteins,inhibit the expression of Bcl-2,Survivin genes and proteins,and promote apoptosis of A549 cells.
