摘  要:目的:分析肾癌组织中miR-193a的表达情况,预测其靶基因,探讨其参与恶性肿瘤调控的分子机制。方法:利用癌症基因组图谱(TCGA)数据库,分析miR-193a在肾透明细胞癌组织与正常组织之间的表达差异。利用Kaplan-Meier plotter数据库,对存在差异表达miR-193a的肾透明细胞癌患者进行生存分析。采用TargetScan7.2、miRDB、PicTar和miRTarBase进行靶基因预测,利用metascape网络在线分析数据库对hsa-miR-193a-3p的靶基因集合进行GO功能注释和KEGG通路富集分析。结果:miR-193a在肾透明细胞癌等多种肿瘤中表达上调,且其表达量与患者总生存时间存在显著负相关。hsa-miR-193a-3p靶基因GO功能主要包括发育过程、细胞成分组织或生物发生、生物过程的正负调节、节律过程等,KEGG信号通路主要富集在肾细胞癌、PI3K-Akt信号通路等方面。结论:hsa-miR-193a-3p可能通过调控多个靶基因参与肾透明细胞癌发生发展相关信号通路的调节,是一个非常具有研究价值的生物学靶标。Objective:To study the expression of miR-193a in renal cellcarcinoma and predict its target genes,so as to explore the molecular mechanism of these genes involved in malignant tumors.Methods:The difference of miR-193a expression between clear cell renal cell carcinoma and normal tissues was analyzed by using the Cancer Genome Atlas(TCGA)database.The Kaplan-Meier curve was used to analyze the survival of clear cell renal cell carcinoma patients with differential expression of miR-193a.Target genes of hsa-miR-193a-3p were predicted by TargetScan 7.2,miRDB,Pictar and miRTarBase databases,GO and KEGG pathway enrichment analysis was performed by using metascape network analysis database.Results:miR-193a was up-regulated in clear cell renal cell carcinoma and other tumors,and there was a negative correlation between miR-193a expression and total survival time.The GO functions of hsa-miR-193a-3p target gene mainly include developmental processes,cellular components,tissues or biogenesis,positive and negative regulation of biological processes,and rhythmic processes.KEGG signal pathway is mainly enriched in renal cell carcinoma and PI3K-Akt signaling pathway.Conclusion:hsa-miR-193a-3p may be involved in the regulation of multiple target genes and in the development of clear cell renal cell carcinoma.It is a very valuable biological target for further investigation.
