摘  要:采用HPLC法测定了大果山楂果实中苹果酸、柠檬酸、酒石酸与抗坏血酸的含量。采用C_(18)色谱柱,以0.10%磷酸溶液∶甲醇体积比(95∶5)为流动相,等度洗脱,流速为0.80 mL/min,柱温为30℃,检测波长为214 nm(苹果酸、柠檬酸、酒石酸)、265 nm(抗坏血酸),进样量为10μL。苹果酸、柠檬酸、酒石酸与抗坏血酸在适量浓度范围内与峰面积的线性关系良好,相关系数(r)分别为0.9997、0.9998、0.9966和0.9998。其重复性试验结果的RSD(n=6)分别为1.4%、1.9%、1.6%和1.8%;平均加样回收率(n=6)分别为96.0%(RSD=1.7%)、103.8%(RSD=1.5%)、97.1%(RSD=0.80%)和96.4%(RSD=0.70%)。本实验建立的含量测定方法,结果准确、灵敏度高,为大果山楂质量控制提供了一种手段。The contents of malic acid,citric acid,tartaric acid and ascorbic acid in fruit of mali fructus were determined by high performance liquid chromatography(HPLC)method.The separation was performed in a C_(18) column with 0.10%phosphoric acid and methanol(volume ratio 95∶5)as mobile phase at the flow rate of 0.80 mL/min.The column temperature was set at 30℃,and the detection wavelength was set at 214 nm(for malic acid,citric acid and tartaric acid),265 nm(for ascorbic acid).The injection volume was 10μL.Malic acid,citric acid,tartaric acid and ascorbic acid had good linear relationship in a certain concentration range and the correlation coefficients were 0.9997,0.9998,0.9966 and 0.9998,respectively.Their RSD(n=6)of reproducibility were 1.4%,1.9%,1.6%and 1.8%respectively.The average recoveries(n=6)were 96.0%(RSD=1.7%),103.8%(RSD=1.5%),97.1%(RSD=0.80%)and 96.4%(RSD=0.70%)respectively.This determination method was accurate and had high sensitivity,which provided a mean for quality control of malifructus.
