摘 要:目的 通过对TCGA测序数据库的挖掘,筛选肝细胞癌(hepatocellular carainoma, HCC)预后相关铁死亡基因并构建Signature预后风险评估模型。方法 从TCGA数据库获取HCC测序数据,采用R语言进行铁死亡相关基因表达差异性分析,采用Log Rank检验和单变量Cox比例风险回归进行K-M生存分析。基于time ROC分析及LASSO Cox回归,构建Signature预后模型。利用时间依赖ROC曲线与AUC对预后风险模型的预测能力进行评估。结果 初步筛选得到21个差异表达的铁死亡相关基因,经单变量Cox比例风险回归和K-M生存分析,进一步筛选得到7个预后相关的铁死亡相关基因。基于上述预后相关铁死亡差异性基因集进行Signature预后模型的构建,lambda.min=0.0317,Risk score=0.118×FANCD2+0.1514×SLC1A5+0.1632×SLC7A11。利用时间依赖ROC曲线与AUC对预后风险模型的预测能力进行评估,结果显示,1年预测效能AUC=0.773,3年AUC=0.667,5年AUC=0.658。结论 通过对TCGA测序数据库的挖掘,筛选并构建基于FANCD2、SLC1A5、SLC7A11三个铁死亡相关差异基因构建的预后风险评估模型具有良好的诊断效能,对HCC预后预测具有重要价值。Objective Based on TCGA sequencing database,hepatocellular carainoma(HCC)prognostic ferroptosis-related genes were screened and Signature prognostic risk assessment model was constructed.[WTHZ]Methods HCC sequencing data were obtained from TCGA database and the differences in ferroptosis-related genes expression were analyzed by R language.K-M survival analysis was performed by Log Rank test and univariate Cox proportional risk regression.Signature prognostic model was established based on time ROC analysis and LASSO Cox regression.Time-dependent ROC curves and AUC were used to evaluate the predictive power of the prognostic risk model.[WTHZ]Results Twenty-one differentially expressed ferroptosis-related genes were initially screened,and seven prognostic ferroptosis-related genes were further screened by univariate Cox proportional risk regression and K-M survival analysis.The Signature prognostic model was constructed based on the above prognostic ferroptosis-related gene set,lambda.min=0.0317,Risk score=0.118×FANCD2+0.1514×SLC1A5+0.1632×SLC7A11.The predictive ability of the prognostic risk model was evaluated by the time-dependent ROC curve and AUC,and the results showed that the one-year predictive efficiency AUC was 0.773 tree-year AUC was 0.667,five-year AUC was 0.658.[WTHZ]Conclusion Through the mining of TCGA sequencing database,the prognostic risk assessment model based on FANCD2,SLC1A5 and SLC7A11 ferroptosis-related differential genes was screened and constructed,which has good diagnostic efficacy and is of great value for the prognosis prediction of HCC.
