摘  要:uge1基因编码一种参与半乳糖代谢的酶,对细胞生长和有性繁殖等有着重要作用.该文以裂殖酵母(Schizosaccharomyces pombe)为材料,采用激光共聚焦活细胞成像的方式首次探究了uge1基因缺失对有丝分裂的分裂期染色体、纺锤体、肌动蛋白动力学的影响.对间期微管的观察结果显示,uge1Δ细胞中含有5根微管的细胞比例增加.有丝分裂期的染色体、纺锤体动力学结果表明,uge1基因缺失会导致染色体分离异常、单极纺锤体的出现、纺锤体从形成到解聚的时间显著增加.uge1基因缺失还会影响裂殖酵母有丝分裂前期和后期纺锤体伸长.纺锤体形成类型显示,uge1Δ细胞中“棒状”纺锤体的比例显著降低,而“点状”纺锤体比例则显著增加.uge1Δ细胞的形态相较野生型细胞有显著变化,表现为长度增加及长宽比增加.肌动蛋白动力学跟踪结果表明,uge1Δ细胞中肌动蛋白束长度、从聚合到解聚的时间、停留时间和收缩时间均显著增加,而肌动蛋白从聚合到解聚的速率显著降低.以上结果表明,uge1基因缺失会导致有丝分裂染色体、纺锤体及肌动蛋白动力学缺陷.The uge1 gene encodes an enzyme involved in galactose metabolism,which plays an important role in cell growth and sexual reproduction.In this study,Schizosaccharomyces pombe was used as a system to analyze the effects of uge1 gene deletion on the dynamics of chromosomes,spindles and actin during mitosis by laser confocal imaging for the first time.Observations on interphase microtubules showed that the percentage of 5 microtubules in uge1Δcells increased compared to wild type cells.Deletion of uge1 gene led to abnormal chromosome segregation,and both the frequency of monopolar spindle and the time between formation and depolymerization for spindle increased significantly.Deletion of uge1 gene also affected the elongation of spindle in mitotic prophase and anaphase.Examination on the type of spindle formation indicated that the frequency of“bar-shaped”spindle in uge1Δcells was significantly reduced,and the“dot-shaped”spindle was significantly increased.Observations on the cell morphology suggested that both the length and ratio of length-width of uge1Δcells increased.Besides,the length,the time from polymerization to depolymerization,the residence time and shrinkage time of actin bundles in uge1Δcells increased significantly,while the rate of actin depolymerization in uge1Δcells reduced significantly.These results indicated that the deletion of uge1 gene would lead to a variety of defects of mitotic chromosome,spindle and actin dynamics.
