摘  要:目的 验证新型VCP/p97抑制剂CB-5083对前列腺癌细胞的抗肿瘤作用,并分析其促进癌细胞凋亡的细胞分子机制。方法 使用呈浓度梯度的CB-5083处理PC-3细胞。采用EdU法监测细胞增殖能力,划痕实验和Transwell法监测细胞迁移、侵袭能力,流式细胞仪检测细胞凋亡,比色法检测Caspase活性,Western Blot检测相关蛋白改变。结果 CB-5083能够抑制PC-3细胞的生长、迁移和侵袭,并促进其凋亡,其作用强弱与药物剂量相关。进一步研究发现CB-5083通过Caspase依赖的内源性途径诱导的细胞凋亡。结论 CB-5083能在前列腺癌细胞中发挥抗肿瘤作用,并通过Caspase依赖的内源性途径诱导前列腺癌细胞凋亡,是一种潜在的治疗策略。Objective To verify the anti-tumor effect of the new VCP/p97 inhibitor CB-5083 on the growth,migrations of prostate cancer(PC)cells and find the cellular molecular mechanism of promoting death among PC patients.Methods PC-3 cells were treated with CB-5083 with concentration gradient.The ability of cell proliferation was detected by EdU method;the ability of cell migration and invasion was detected by scratch test and Transwell method;apoptosis was detected by flow cytometry;the activity of Caspase was detected by colorimetry,and the changes of related proteins were detected by Western Blot.Results CB-5083 could inhibit the growth,migration and invasion of PC-3 cells and promote its apoptosis,and its effect was positively correlated with the drug dose.Further study found that CB-5083 induced apoptosis through Caspase-dependent endogenous pathway.Conclusions CB-5083 can exert anti-tumor effect in prostate cancer cells and induce apoptosis of PC cells through Caspase-dependent endogenous pathway.CB-5083 treatment of PC will be a potential therapeutic strategy.
