摘　　要：目的 探讨叉头框E1蛋白(FOXE1)基因对结直肠癌细胞迁移能力的影响。方法 将人结直肠癌细胞株RKO分为实验组(RKO细胞转染FOXE1表达质粒)和对照组(RKO细胞转染空载质粒),采用半定量RT-PCR法分析FOXE1 mRNA在结直肠癌细胞株中的表达量,蛋白免疫印迹检测实验组和对照组细胞中FOXE1蛋白及细胞骨架相关蛋白的表达量。通过划痕实验、Transwell实验和细胞免疫荧光实验观察FOXE1对肿瘤细胞迁移能力和细胞形态的影响。结果 FOXE1基因在结直肠癌细胞株中表达沉默,结直肠癌细胞株转染FOXE1后,RKO细胞的划痕愈合能力和细胞迁移能力明显降低,细胞伪足形成明显减少,高表达FOXE1的RKO细胞切丝蛋白表达降低,而磷酸化失活的切丝蛋白表达升高。结论 FOXE1基因通过磷酸化失活切丝蛋白影响细胞肌动蛋白骨架重组以及丝状伪足形成,从而抑制结直肠癌细胞的迁移。Objective To investigate the effect of fbrkhead box El protein(FOXE1)gene on the migration ability of colorectal cancer cell migration.Methods Human colorectal cancer cell line RKO was divided into experimental group(RKO cells transfected with FOXE1 expression plasmid)and control group(RKO cells transfected with empty plasmid).The expression of FOXE 1 mRNA in colorectal cancer cell lines was analyzed by semi quantitative RT-PCR,and the expression of FOXE 1 protein and cytoskeleton related protein in experimental and control groups were detected by Western Blot assay.The effects of FOXE 1 on tumor cell migration and cell morphology were observed by scratch test,Transwell test and cellular immunofluorescence test.Results FOXE 1 was silenced in colorectal cancer cell lines.After transfection with FOXE1,the ability of wound healing and migration ofRKO cells were significantly inhibited,and the formation of pseudopodia was significantly reduced.The expression of cofilin in RKO cells with high FOXE1 expression was decreased,while the expression of phospho-Cofilin was increased.Conclusions FOXE1 may inhibit the migration of colorectal cancer cells by affecting cytoskeleton reorganization and pseudopodia formation due to cofilin phosphorylation.
