核仁素定位和定量表达与鼻咽癌演变的关系

摘　　要：目的:发现核仁素在原发性鼻咽癌、鼻咽部上皮内瘤变及正常上皮组织中定位和定量表达的差异,探讨核仁素在鼻咽癌演变过程中的意义。方法:收集台山市人民医院外检病例,选择原发性鼻咽癌患者50例、鼻咽部上皮内瘤变患者25例和鼻咽部正常上皮患者25例,采用免疫组化SP法检测鼻咽癌、鼻咽部上皮内瘤变及正常上皮组织中核仁素表达的差异,分析核仁素的定位定量表达在鼻咽癌演变过程中的作用和意义。结果:原发性鼻咽癌组织中核仁素的表达量,显著高于鼻咽部上皮内瘤变组织与鼻咽部正常上皮组织,P<0.05,且鼻咽部上皮内瘤变组织中核仁素的表达量显著高于鼻咽部正常上皮组织,P<0.05。鼻咽癌、鼻咽部上皮内瘤变和正常上皮组织的细胞核中均表达核仁素,鼻咽癌细胞质膜表达核仁素49例,明显高于鼻咽部上皮内瘤变8例及鼻咽部正常上皮1例,P<0.05。结论:核仁素在鼻咽癌的发生发展中呈升高表达趋势,同时可引起核外迁移,是评估鼻咽癌演变的重要蛋白标志物。Objective:To investigate the difference of localization and quantitative expression of nuclein in primary nasopharyngeal carcinoma,nasopharyngeal intraepithelial neoplasia and normal epithelial tissue,and to explore the significance of nuclein in the evolution of nasopharyngeal carcinoma.Method:A total of 50 patients with primary nasopharyngeal carcinoma,25 patients with nasopharyngeal intraepithelial neoplasia and 25 patients with nasopharyngeal normal epithelium were selected for external examination in Taishan People’s Hospital.SP immunohistochemical method was used to detect the differences of nucleolin expression in nasopharyngeal carcinoma,nasopharyngeal intraepithelial neoplasia and normal epithelial tissues.To analyze the role and significance of localization and quantitative expression of nuclein in the evolution of nasopharyngeal carcinoma.Results:The expression level of nuclein in primary nasopharyngeal carcinoma tissues was significantly higher than that in nasopharyngeal intraepithelial neoplasia tissues and normal nasopharyngeal epithelial tissues(P<0.05),and the expression level of nuclein in nasopharyngeal intraepithelial neoplasia tissues was significantly higher than that in normal nasopharyngeal epithelial tissues(P<0.05).Nucleonin was expressed in the nuclei of nasopharyngeal carcinoma,nasopharyngeal intraepithelial neoplasia and normal epithelial tissues.The plasma membrane of nasopharyngeal carcinoma cells expressed nucleonin in 49 cases,which was significantly higher than that of nasopharyngeal intraepithelial neoplasia in 8 cases and normal epithelium in 1 case,P<0.05.Conclusion:The expression of nuclein is increased in the development and progression of nasopharyngeal carcinoma,and it can also cause extracellular migration,which is an important protein marker to evaluate the evolution of nasopharyngeal carcinoma.
