摘 要:目的建立肺腺癌SPC-A1细胞裸鼠移植瘤模型,探讨毛蕊异黄酮对肺腺癌移植瘤生长及凋亡的影响。方法30只裸鼠(6~8周龄)适应性饲养一周后,将制备好的SPC-A1细胞悬液(浓度为5×105个/mL)经腋下皮下注射,拟建造SPC-A1肺腺癌细胞移植瘤模型;按照裸鼠的重量及瘤体的体积大小,随机分为模型组、毛蕊异黄酮组、顺铂组,每组6只;记录各组裸鼠实验前后的体质量、瘤重、瘤体横径和纵径,并分析各组裸鼠模型的肿瘤抑制率,测估SPC-A1裸鼠移植瘤的体积大小及生长状况;通过末端脱氧核苷酸转移酶介导的脱氧尿苷三磷酸缺口末端标记(TUNEL)测定法测定SPC-A1细胞裸鼠移植瘤的凋亡状况;蛋白免疫印迹法(Western blot)测定各组瘤体中凋亡相关蛋白Bax、Bcl-2的表达水平。结果顺利构建了肺腺癌SPC-A1细胞裸鼠移植瘤的模型。与模型组比较,毛蕊异黄酮组裸鼠瘤的瘤重[(1.20±0.19)g vs(0.98±0.14)g]明显降低,差异有统计学意义(P<0.05),且抑瘤率为18.33%,表明毛蕊异黄酮能有效抑制SPC-A1细胞裸鼠移植瘤的生长;TUNEL法检测的各组裸鼠移植瘤细胞的凋亡结果分析表明,与模型组比较,毛蕊异黄酮组凋亡率[(10.00±0.82)%vs(32.43±1.99)%]明显增加,差异有统计学意义(P<0.05),证实毛蕊异黄酮可有效诱导肺腺癌SPC-A1细胞的凋亡作用;Western blot结果显示,毛蕊异黄酮能明显上调促凋亡相关蛋白Bax的表达水平,同时能明显下调抑凋亡相关蛋白Bcl-2的表达水平,差异有统计学意义(P<0.05)。结论毛蕊异黄酮能够明显抑制SPC-A1裸鼠移植瘤的生长并促进肺癌细胞的凋亡。Objective To establish transplanted tumor models of lung adenocarcinoma cell SPC-A1 in nude mice and investigate the effect of calycosin on the proliferation and apoptosis of transplanted tumor of lung adenocarcinoma.Methods After the adaptive feeding of 30 nude mice(6 to 8 weeks old)for one week,the prepared SPC-A1 cell suspension(at a concentration of 5×105 cells/mL)was injected under the armpit to establish a SPC-A1 lung cancer cell transplanted model.According to the weight of nude mice and the volume size of the tumor body,the mice were randomly divided into model group,calycosin group,and cisplatin group,with six mice in each group.The weight,tumor weight,transverse diameter,and longitudinal diameter of nude mice were recorded to analyze the tumor suppression rate of each nude mouse model and assess the tumor size and growth of SPC-A1 nude mice.The apoptosis of SPC-A1 cells in transplanted nude mice was determined by Terminal deoxynucleotide transferase-mediated dUTP nick-end labeling(TUNEL).The expression of the apoptosis-related proteins Bax and Bcl-2 in each group was determined through western blot.Results The transplanted tumor models of SPC-A1 cells in lung adenocarcinoma were successfully constructed.In comparison with the model group,the tumor weight in calycosin group was decreased significantly:(1.20±0.19)g vs(0.98±0.14)g,P<0.05,and the tumor suppression rate reached 18.33%,which indicated that calycosin can effectively inhibit the growth of SPC-A1 cell transplanted tumors in nude mice.TUNEL showed that the apoptosis rate of the transplanted tumor cells was increased significantly in the calycosin group than the model group:(10.00±0.82)%vs(32.43±1.99)%,P<0.05,which conformed that calycosin can effectively induce the apoptosis of SPC-A1 cells in lung adenocarcinoma.Western blot results showed that calycosin can significantly up-regulate the expression level of the apoptosis-related protein Bax and also significantly down-regulate the expression level of the apoptosis-related protein Bcl-2(P<0.
